败酱草总皂苷含药血清对人胃癌SGC-7901细胞增殖的影响及机制研究

摘　　要：目的探讨败酱草总皂苷含药血清对人胃癌SGC-7901细胞增殖和细胞周期的影响。方法取购自中国科学院典型培养物保藏委员会细胞库的人胃癌SGC-7901细胞株进行培养,分为对照组和药物组,对照组加入含10%澳洲胎牛血清的RPMI1640培养液,药物组按加入败酱草总皂甙含药血清浓度分为低浓度组、中浓度组和高浓度组,采用CCK8法检测各组细胞活力,采用流式细胞术检测各组细胞周期,采用Western blot技术检测各组细胞周期相关蛋白[细胞周期蛋白D1(CyclinD1)、细胞周期蛋白E(CyclinE)、细胞周期蛋白依赖性激酶4(CDK4)、自噬核心蛋白(Beclin-1)]表达水平。结果CCK8法检测结果显示,药物组培养24、48、72h的OD值均低于对照组(P<0.05),OD值高浓度组<中浓度组<低浓度组(P<0.05);流式细胞术检测结果显示,与对照组比较,药物组细胞G1期细胞百分比明显升高(P<0.05),G2期、S期细胞百分比明显降低(P<0.05),细胞百分比随着药物血清浓度升高、降低而增加、减少;Western blot技术检测结果显示,与对照组比较,药物组细胞周期相关蛋白CyclinD1、CyclinE、CDK4表达水平明显降低(P<0.05),高浓度组<中浓度组<低浓度组;Beclin-1蛋白表达水平明显升高(P<0.05),高浓度组>中浓度组>低浓度组。结论败酱草总皂苷含药血清可抑制人胃癌SGC-7901细胞增殖,阻滞细胞周期于G1期,其机制可能与调节细胞周期相关蛋白表达有关。Objective To explore the effects of total saponins of patrinia containing serum on proliferation and cell cycle of human gastric cancer SGC-7901 cells.Methods Human gastric cancer SGC-7901 cell lines purchased from cell bank of typical culture preservation committee of the Chinese Academy of Sciences were cultured and divided into control group and drug group.Control group was supplemented with RPMI1640 culture solution containing 10% Australian fetal bovine serum,and drug group was divided into low concentration group,medium concentration group and high concentration group according to the concentration of total saponins of patrinia containing serum.CCK8 assay was used to detect the cell viability in each group.Flow cytometry was used to detect the cell cycle of each group.Western blot assay was used to detect the expression levels of cell cycle-related proteins[CyclinD1,CyclinE,Cyclin-dependent kinase4(CDK4),Beclin-1].Results CCK8 assay showed that the OD values in drug group at 24,48 and 72h of culture were lower than those in control group(P<0.05),and the OD value showed high concentration groupmedium concentration group>low concentration group.Conclusion Total saponins of patrinia containing serum can inhibit the proliferation of human gastric cancer SGC-7901 cells,block the cell cycle in G1phase,and the mechanism may be related to the regulation of cell cycle-related protein expression.
