环指蛋白187在乳癌组织中的表达及其临床意义

摘　　要：目的 探讨环指蛋白187(RNF187)在乳癌组织中的表达及其与临床病理特征的关系。方法 选择行乳癌改良根治术的女性病人52例,其中发生间质脉管癌栓29例(高风险组),没有发生间质脉管癌栓23例(低风险组)。采用实时定量PCR法和免疫组织化学法,检测各组乳癌及癌旁正常组织中RNF187 mRNA和蛋白的表达,并分析其表达与病人临床病理特征的关系。结果 乳癌组织中RNF187 mRNA的表达明显高于癌旁正常组织,差异有统计学意义(t=2.887,P<0.05);高风险组RNF187 mRNA表达明显高于低风险组(t=3.676,P<0.05)。RNF187阳性染色主要定位于肿瘤细胞的胞质及胞核中,尤以胞质为主,乳癌组织呈明显深染,癌旁正常组织呈淡染,乳癌组织中RNF187表达与相应癌旁正常组织比较差异有统计学意义(χ^(2)=22.190,P<0.01)。高风险组RNF187表达与低风险组比较差异有统计学意义(χ^(2)=4.293,P<0.05)。结论 RNF187在乳癌组织中的高表达可能与乳癌的易复发、转移等不良预后有关。RNF187可能成为判断乳癌不良预后的指标及临床治疗的靶点。Objective To investigate the expression of ring finger protein 187(RNF187) in breast cancer and its association with clinicopathological features. Methods A total of 52 female patients who underwent modified radical mastectomy for breast cancer were enrolled, among whom there were 29 patients with interstitial vascular tumor thrombus(high-risk group) and 23 patients without interstitial vascular tumor thrombus(low-risk group). Quantitative real-time PCR and immunohistochemistry were used to measure the mRNA and protein expression of RNF187 in breast cancer tissue and adjacent tissue in each group, and the association between RNF187 expression and clinicopathological features was analyzed. Results The mRNA expression of RNF187 in breast cancer tissue was significantly higher than that in adjacent tissue(t=2.887,P<0.05), and the high-risk group had a significantly higher mRNA expression of RNF187 than the low-risk group(t=3.676,P<0.05). RNF187 positive staining was mainly localized in the cytoplasm and nuclei of tumor cells, especially in the cytoplasm;breast cancer tissue showed obvious deep staining, while normal adjacent tissue showed light staining. There was a significant difference in the expression of RNF187 between breast cancer tissue and normal adjacent tissue(χ^(2)=22.190,P<0.01), and there was also a significant difference in the expression of RNF187 between the high-risk group and the low-risk group(χ^(2)=4.293,P<0.05). Conclusion The high expression of RNF187 in breast cancer may be associated with the poor prognosis of breast cancer, such as frequent recurrence and metastasis. RNF187 may become an indicator for poor prognosis of breast cancer and a target for clinical treatment.
