摘 要:目的 研究葛根素、姜黄素、沙棘黄酮复方对急性四氯化碳肝损伤的保护作用。方法 将60只雄性昆明种小鼠按体质量随机分为对照组(Control)、模型组(Model)、葛根素组(Pue)、姜黄素组(Cur)、沙棘黄酮组(Seaf)和复方组(Pue+Cur+Seaf)。各组小鼠连续灌胃给药15 d后,以40%CCl40.4 mL制作小鼠肝损伤模型,16 h后测定各组小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,肝匀浆丙二醛(MDA)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)、甘油三酯(TG)、总胆固醇(TCHO)含量并进行肝脏病理组织学检查。结果 与模型组相比,复方组小鼠血清中ALT、AST活性升高的趋势下降;小鼠肝匀浆MDA、TG含量显著低于模型组,SOD活性、GSH均显著高于模型组;TCHO含量与模型组比较,差异无统计学意义。以上结果及病理组织学检查显示复方组各项指标多数优于其他单一材料作用组。结论 葛根素、姜黄素、沙棘黄酮均能不同程度降低四氯化碳造成的肝损伤,三者单独作用效果姜黄素相对较好,而三者以1∶1∶1比例混合组成的复方组更显著的降低四氯化碳造成的肝损伤,对小鼠肝损伤具有一定辅助保护作用。Objective:To study the protective effects of puerarin,curcumin and sea-buckthorn flavone compound on acute carbon tetrachloride liver injury.Methods:60 male Kunming mice were randomly divided into control group(Control),model group(Model),Puerarin group(Pue),curcumin group(Cur),seabuckine flavonoid group(Seaf)and compound group(Pue+Cur+Seaf)according to body weight.After 15 days of continuous intragastric administration,the mice in each group were treated with 40%CCl40.4 mL to make liver injury model.The activities of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum of mice in each group were measured 16 h later.The contents of malondialdehyde(MDA),superoxide dismutase(SOD),reduced glutathione(GSH),triglyceride(TG)and total cholesterol(TCHO)in liver homogenate were examined by histopathology.Results:Compared with that of the model group,the activity of ALT and AST in serum of compound group decreased.The contents of MDA and TG in mouse liver homogenate were significantly lower than those in the model group,while SOD activity and GSH were significantly higher than those in the model group.There was no statistical difference when compared TCHO content with that of the model group.The above results and histopathological examination showed that most of the indexes of the compound group were better than those of other single material groups.Conclusions:Puerarin,curcumin and sea-buckrhamnoflavone can all reduce the liver injury caused by carbon tetrachloride to varying degrees.The effect of curcumin alone is better than those of puerarin and sea-buckrhamnoflavone while the compound group composed of the three at the ratio of 1∶1∶1 can significantly reduce the liver injury caused by carbon tetrachloride,and has a certain adjuvant protective effect on the liver injury of mice.
