摘　　要：目的研究长链非编码RNA AC098934对非小细胞肺癌细胞侵袭性的影响和机制。方法Transwell实验检测转染siRNA的人非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞系A549及H1299,检测其侵袭性的变化;转染lncRNA AC098934于A549及NCI-H1299细胞中,将其分为3组,分别为NC组、OE-AC098934组及OE+SB203580组,通过Transwell实验检测其侵袭性的变化。Western Blot检测抑制lncRNA AC098934的A549及NCI-H1299细胞样本中p38、p-p38、JNK、p-JNK、Myc、p53蛋白的表达水平。结果抑制lncRNA AC098934的表达,可明显抑制A549及H1299细胞的侵袭性。lncRNA AC098934过表达后,A549及NCI-H1299细胞侵袭性增强(P<0.05),而抑制p38MAPK通路后,A549及NCI-H1299细胞侵袭性受到抑制。抑制lncRNA AC098934表达的A549及NCI-H1299细胞,其Myc、p-p38/p38、p-JNK/JNK蛋白表达提高(P<0.05),p53蛋白降低(P<0.05)。结论lncRNA AC098934通过激活p38MAPK通路增强NSCLC细胞的侵袭性。Objective To investigate the effect and mechanism of long non-coding RNA AC098934 on the invasiveness of human non-small-cell lung cancer(NSCLC).Methods Transwell assay was used to detect the changes of invasiveness for NSCLC cell line A549 and H1299 after being transfected with si-RNA.LncRNA AC098934 was transfected into A549 and NCI-H1299 cells,and they were divided into 3 groups,namely NC group,OE-AC098934 group and OE+SB203580 group.Their changes of invasiveness were detected by Transwell assay.Western Blot was used to detect the protein expressions of p38,p-p38,JNK,p-JNK,Myc and p53 in A549 and NCI-H1299 cells which inhibited lncRNA AC098934.Results Inhibiting the expression oflncRNA AC098934 could significantly reduce the invasiveness of A549 and H1299 cells.The invasiveness of A549 and NCI-H1299 cells was enhanced(P<0.05)with overexpression of lncRNA AC098934,while the invasiveness of A549 and NCI-H1299 cells was inhibited after the inhibition of p38MAPK pathway.The protein expressions of Myc,p-p38/p38 and p-JNK/JNK were increased(P<0.05)and p53 protein expression decreased(P<0.05)when the expression lncRNA AC098934 of A549 and NCI-H1299 cells was inhibited.Conclusion LncRNA AC098934 enhances the invasiveness of NSCLC cells by activating the p38MAPK pathway.
