摘　　要：目的:探讨微小RNA(miRNA,miR)-365在胆囊癌中的表达及其对肿瘤进展的影响。方法:选取2019年6月到2021年6月商丘市第一人民医院收治的53例胆囊癌患者的肿瘤组织和癌旁组织作为研究对象,采用荧光定量聚合酶链反应(PCR)分析胆囊癌组织和癌旁组织中miR-365表达水平。以人胆囊癌细胞株GBC-SD为研究对象,转染对照miRNA和miR-365模拟物,建立对照组和观察组细胞系,采用细胞计数试剂盒(CCK-8)分析细胞增殖活力,采用体外移植瘤实验分析细胞体内增殖能力,采用流式细胞术分析细胞凋亡比例,采用Transwell分析细胞的迁移能力,采用蛋白质印迹法(Western blot)分析细胞上皮-间充质转化能力和凋亡相关蛋白表达。组间计量数据比较采用t检验。结果:癌旁组织中miR-365表达水平(0.92±0.12)明显高于胆囊癌组织(0.54±0.16),差异有统计学意义(t=14.001,P<0.05)。对照组细胞吸光度(A)值(1.90±0.12)明显高于观察组(1.26±0.17),差异有统计学意义(t=7.737,P<0.05)。对照组细胞肿瘤体积[(666.33±66.97)mm 3]明显高于观察组[(399.17±46.23)mm 3],差异有统计学意义(t=8.041,P<0.05)。对照组细胞肿瘤质量[(3.56±0.45)g]明显高于观察组[(2.01±0.11)g],差异有统计学意义(t=8.236,P<0.05)。对照组细胞凋亡比例[(3.48±0.53)%]明显低于观察组[(25.25±3.51)%],差异有统计学意义(t=15.080,P<0.05)。Transwell结果显示,对照组细胞迁移数量[(89.67±8.29)个]明显低于观察组[(42.33±9.11)%],差异有统计学意义(t=9.412,P<0.05)。对照组细胞上皮细胞标志物E-钙黏蛋白(E-cadherin)蛋白表达水平(1.08±0.08)明显低于观察组(1.99±0.15),差异有统计学意义(t=12.850,P<0.05)。对照组细胞角形蛋白(Vimentin)和N-钙黏蛋白(N-cadherin)蛋白表达水平(1.08±0.07、1.50±0.11)明显高于观察组(0.71±0.05、0.69±0.12),差异有统计学意义(t=10.420、11.880,P<0.05)。对照组细胞第10号染色体上缺失与张力蛋白同源的磷酸�Objective To investigate the expression of microRNA(miRNA,miR)-365 in gallbladder carcinoma and its effect on tumor progression.Methods The tumor tissues and adjacent tissues of 53 patients with gallbladder cancer admitted to our hospital from June 2019 to June 2021 were selected as the research objects.The expression level of miR-365 in gallbladder cancer tissues and adjacent tissues was analyzed by fluorescence quantitative polymerase chain reaction(PCR).Human gallbladder cancer cell line GBC-SD was used as the research object.Control miRNA and miR-365 mimics were transfected to establish cell lines in the control group and the observation group.Cell proliferation activity was analyzed by cell counting kit-8(CCK-8)and cell proliferation ability in vivo was analyzed by in vitro tumor transplantation experiment.The ability of epithelial mesenchymal transition and the expression of apoptosis related proteins were analyzed by Western blotting.T test was used to compare the measurement data between groups.Results The expression level of miR-365 in adjacent tissues(0.92±0.12)was significantly higher than that in gallbladder cancer tissues(0.54±0.16,t=14.001,P<0.05).The absorbance(A)value of cells in the control group(1.90±0.12)was significantly higher than that of cells in the observation group(1.26±0.17,t=7.737,P<0.05).The cell tumor volume of the control group[(666.33±66.97)mm3]was significantly higher than that of the observation group[(399.17±46.23)mm3,t=8.041,P<0.05].The weight of cell tumor in the control group[(3.56±0.45)g]was significantly higher than that in the observation group[(2.01±0.11)g,t=8.236,P<0.05].The percentage of apoptosis in the control group[(3.48±0.53)%]was significantly lower than that in the observation group[(25.25±3.51)%,t=15.080,P<0.05].The number of cell migration in the control group(89.67±8.29)was significantly lower than that in the observation group[(42.33±9.11)%,t=9.412,P<0.05].The expression level of E-cadherin protein,a marker of epithelial cells,in the control group(
