摘  要:目的 探讨泛素结合酶E2T(UBE2T)对鼻咽癌细胞放疗敏感性的影响及相关分子机制。方法 采用qRT-PCR检测鼻咽癌细胞CNE2及正常鼻咽上皮细胞NP69中UBE2T的表达水平。将UBE2T小干扰RNA序列(UBE2T-siRNA)及其阴性对照序列(NC-siRNA)转染至CNE2细胞,联合或不联合放射治疗,并分为Control组、NC-siRNA组、UBE2T-siRNA组、放疗组、NC-siRNA+放疗组、UBE2T-siRNA+放疗组。采用CCK-8检测各组细胞增殖能力;流式细胞术检测细胞凋亡情况;Western blot检测Wnt/β-catenin信号通路相关蛋白表达水平。结果 CNE2细胞中UBE2T mRNA表达水平高于NP69细胞(P<0.05);与Control组比较,NC-siRNA组细胞中UBE2T mRNA表达水平无明显变化(P>0.05),UBE2T-siRNA组细胞中UBE2T mRNA表达水平显著降低(P<0.05)。与Control组比较,放疗组、UBE2T-siRNA组细胞增殖活性均降低,细胞凋亡率均升高(P<0.05);与放疗组和UBE2T-siRNA组比较,UBE2T-siRNA+放疗组细胞增殖活性降低,细胞凋亡率升高(P<0.05)。与Control组比较,放疗组、UBE2T-siRNA组细胞中Wnt3α、β-catenin、c-myc蛋白表达均降低(P<0.05);与放疗组和UBE2T-siRNA组比较,UBE2T-siRNA+放疗组细胞中Wnt3α、β-catenin、c-myc蛋白表达均降低(P<0.05)。结论 UBE2T通过调控Wnt/β-catenin信号通路增强鼻咽癌细胞的放疗敏感性。Objective To investigate the effect of ubiquitin-conjugating enzyme E2T(UBE2T) on radiosensitivity of nasopharyngeal carcinoma cells and its related molecular mechanism.Methods The expression levels of UBE2T in nasopharyngeal carcinoma cell CNE2 and normal nasopharyngeal epithelial cell NP69 were detected by qRT-PCR.UBE2T small interfering RNA sequence(UBE2T-siRNA) and its negative control sequence(NC-siRNA) were transfected into CNE2 cells with or without radiotherapy and the cells were divided into the Control group, the NC-siRNA group, the UBE2T-siRNA group, the radiotherapy group, the NC-siRNA+radiotherapy group and the UBE2T-siRNA+radiotherapy group. The cell proliferation was detected by CCK-8,the cell apoptosis was detected by flow cytometry, and the expression levels of Wnt/β-catenin signaling pathway related proteins were detected by Western blot.Results The expression level of UBE2T mRNA in CNE2 cells was higher than that in NP69 cells(P<0.05).Compared with the Control group, the expression level of UBE2T mRNA of cells in the NC-siRNA group had no significant change(P>0.05),while the expression level of UBE2T mRNA of cells in the UBE2T-siRNA group was decreased significantly(P<0.05).Compared with the Control group, the proliferation activities of cells in the radiotherapy group and the UBE2T-siRNA group were decreased and the cell apoptosis rates were increased(P<0.05).Compared with the radiotherapy group and the UBE2T-siRNA group, the cell proliferation activity of cells in the UBE2T-siRNA+radiotherapy group was decreased and the cell apoptosis rate was increased(P<0.05).Compared with the Control group, the expression of Wnt3α,β-catenin and c-myc protein of cells in the radiotherapy group and the UBE2T-siRNA group were decreased(P<0.05);compared with the radiotherapy group and the UBE2T-siRNA group, the expression of Wnt3α,β-catenin and c-myc protein of cells in the UBE2T-siRNA+radiotherapy group was decreased(P<0.05).Conclusion UBE2T enhances the radiosensitivity of nasopharyngeal carcinoma cells
