三黄导赤散联合针刺疗法治疗大鼠口腔溃疡的实验研究

摘　　要：目的:评价中药联合针刺疗法对大鼠口腔溃疡的治疗作用。方法:采用抗原乳化液方法建立大鼠口腔溃疡模型,三黄导赤散灌胃联合针刺足三里、合谷两穴位进行治疗,分为正常组、对照组、中药治疗组、针刺治疗组、中药+针刺治疗组。通过测量溃疡的面积,检测炎症因子白细胞介素-2(interleukin-2,IL-2)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的水平以及外周血中细胞免疫相关CD3^(+)、CD4^(+)、CD8^(+)细胞水平、CD4^(+)/CD8^(+)比值评价治疗效果。结果:中药、针刺治疗可显著促进大鼠口腔溃疡愈合,中药、针刺治疗可显著降低炎性相关因子IL-6、TNF-α水平,升高IL-2水平,并升高免疫相关CD3^(+)、CD4^(+)细胞水平及CD4^(+)/CD8^(+)比值,降低CD8^(+)细胞水平,以上作用针药联合治疗较中药、针刺单一治疗方式效果显著。结论:针药联合治疗效果优于针刺或中药单一治疗的方式,三黄导赤散联合针刺足三里、合谷穴可有效减轻大鼠口腔溃疡炎症反应、促进溃疡的愈合。Objective:To evaluate the therapeutic effect of acupuncture and medicine combined with internal and external administration on oral ulcer in rats.Methods:The oral ulcer model of rats was established by antigen emulsion method.Sanhuangdaochi San was given ingastric administration combined with acupuncture at Zusanli and Hegu acupoints,and the rats were divided into normal group,control group,traditional Chinese medicine(TCM)treatment group,acupuncture treatment group and TCM+acupuncture treatment group.The area of ulcer was measured,inflammatory cytokines levels of interleukin-2(IL-2),IL-6 and tumor necrosis factor-α(TNF-α)were detected.The cellular immune-related CD3+,CD4^(+),CD8^(+)cells levels in peripheral blood and the CD4^(+)/CD8^(+)ratio were evaluated to evaluate the therapeutic effect.Results:TCM and acupuncture treatment promoted oral ulcer healing in rats significantly.TCM and acupuncture treatment can significantly reduced the levels of IL-6 and TNF-α,increased IL-2 level,CD3+and CD4^(+)cells and CD4^(+)/CD8^(+)ratio,and reduced CD8^(+)cells level.Compared with TCM and acupuncture,the combined treatment of acupuncture and Chinese medicine has a significant effect.Conclusion:The combined treatment of acupuncture and medicine is better than acupuncture or TCM single treatment,Sanhuangdaochi San combined with acupuncture Zusanli and Hegu acupoints reduced the inflammatory response effectively and promoted healing of oral ulcer in rats.
