结肠癌组织和癌旁组织MiR-33a-5p表达及临床意义

摘　　要：[目的]探讨结肠癌组织和癌旁组织MiR-33a-5p表达及临床意义。[方法]采用回顾性研究方法,选取78例结肠癌手术患者癌组织标本与癌旁组织标本,采用RT-PCR方法检测组织中MiR-33a-5p表达情况,调查患者的临床资料并进行相关性分析。[结果]癌组织的MiR-33a-5p相对表达水平显著高于癌旁组织(P<0.05)。在癌组织的MiR-33a-5p表达程度与患者的年龄、性别、肿瘤位置、病理类型等无明显相关性(P>0.05),但与临床分期、肿瘤大小、浆膜浸润程度、分化程度等有明显的差异(P<0.05)。相关性Logistic分析结果表明临床分期、肿瘤大小、浆膜浸润是影响MiR-33a-5p表达程度的主要因素(P<0.05)。癌组织中MiR-33a-5p高表达63例,高表达率为80.77%(63/78);癌旁组织MiR-33a-5p高表达13例,高表达率为16.67%(13/78)。癌组织MiR-33a-5p的高表达率明显高于癌旁组织(P<0.001)。患者总体3年存活率为29.5%,MiR-33a-5p高表达患者、MiR-33a-5p低表达患者3年存活率分别为20.6%(13/63)、66.7%(10/15),两者比较差异有统计学意义(P<0.01).[结论]结肠癌患者癌组织中MiR-33a-5p呈现高表达,且与患者临床分期、肿瘤大小、浆膜浸润、分化程度等临床病理特征显著相关,MiR-33a-5p有望成为结肠癌诊断和预后的分子标记物。[Objective]To investigate the expression and clinical significance of MiR-33 a-5 p in colon cancer tissues and adjacent tissues.[Methods]A retrospective study was conducted to select 78 tissue samples from colon cancer surgery patients(lesion group)and adjacent tissue samples(adjacent cancer group).RT-PCR was used to detect the expression of Mir-33 a-5 p in the tissues, and the clinical data of the patients were investigated and the correlation was analyzed.[Results]The relative expression of MiR-33 a-5 p in the lesion group were significantly higher than that in the paracancerous group(P<0.05).In the lesion group, the relative expression level of MiR-33 a-5 p were not correlated with the age, sex, tumor location and pathological type of the patients(P>0.05),and were correlated with clinical stage, tumor diameter, serosal invasion, histological differentiation(P<0.05).Logistic analysis showed that clinical stage, tumor diameter, and serosal infiltration were the main factors affected the relative expression of MiR-33 a-5 p in patients(P<0.05).MiR-33 a-5 p was highly expressed in 63 cases of colon cancer tissues, with a high expression rate of 80.77%(63/78);MiR-33 a-5 p was highly expressed in 13 cases in paracancerous tissues, with a high expression rate of 16.67%(13/78).The high expression rate of MiR-33 a-5 p in colon cancer tissue was significantly higher than that in normal tissues adjacent to the cancer, and the difference was statistically significant(P<0.001).The overall 3-year survival rate of patients was 29.5%.The 3-year survival rates of patients with high MiR-33 a-5 p expression and those with low MiR-33 a-5 p expression were 20.6%(13/63)and 66.7%(10/15),respectively.There statistical significance(P<0.01).[Conclusion]MiR-33 a-5 p are highly expressed in colon cancer patients, which are significantly correlated with clinical pathological features such as clinical stage, tumor diameter, serosal invasion and histological differentiation.MiR-33 a-5 p is promising as a molecular marker for colon cancer dia
