摘 要:目的:通过筛选与抗阻训练效果相关表型指标存在关联的SNP构建遗传易感分数(Genetic Predisposition Scores,GPS)预测久坐汉族青年抗阻训练增肌效果,为精准化运动方案的制定提供依据。方法:187名久坐汉族青年完成12周抗阻训练(男性占比:51.47%),联合股四头肌中段厚度(Quadriceps Muscle Thickness,THK-QUAD)和全身瘦体重(lean-body mass,LBM)变化评测增肌效果,文献资料筛选与抗阻训练相关的基因多态性(Single Nucleotide Polymorphism,SNP),采用Illumina CGA-24v1-0芯片完成基因分型,采用Plink1.09软件的Logistics回归分析筛选出与抗阻训练增肌效果相关联的SNP,进一步采用SAS9.3软件分析与构建GPS模型。结果:文献报道的57个SNPs中有6个(rs7975232、rs6552828、rs768539、rs1800779、rs1024610、rs10263647)被验证与中国汉族青年抗阻训练增肌效果关联(P<0.05),生物信息学分析表明6个SNPs影响36种基因的表达,在趋化因子受体通路、细胞活化以及细胞稳态维持等功能中发挥作用。构建GPS模型后采用ROC曲线和logistics回归分析发现GPS可以解释抗阻训练增肌效果的差异的12.4%,评分大于32分的个体在抗阻训练后取得高增肌效果的概率是低分组的4.27倍(P<0.01)。结论:通过6个SNP构建的GPS模型可以用于久坐汉族青年人抗阻训练增肌效果的预测,GPS大于32分的久坐汉族青年可能在抗阻训练中取得更好的增肌效果。Objective:This research aims to predict the muscle-building effect of resistance training in sedentary Han youths by constructing a Genetic Predisposition Scores(GPS)model.Methods:187(male:51.47%)sedentary Han Youths completed 12 weeks of resistance training.Thickness Quadriceps(THK-QUAD)and lean-body mass(LBM)were used to evaluate the effect of muscle gain.The literature data were screened for gene polymorphisms related to resistance training and genotyped by Illumina CGA-24v1-0 gene chip.The Logistics regression analysis using Plink1.09 software screened out the SNPs associated with the muscle-building effect of resistance training,and SAS9.3 software is further used to analyze and build GPS models.Results:six of the 57 SNPs reported in the literature(rs7975232,rs6552828,rs768539,rs1800779,rs1024610,rs10263647)were verified to be associated with the muscle-building effect of resistance training in Han youths(P<0.05).Bioinformatics analysis showed that 6 SNPs affected the expression of 36 genes.Enrichment analysis found that this played a role in the chemokine receptor pathway,cell activation and maintenance of cell homeostasis.After the GPS model was constructed,ROC and logistics regression analysis were adopted to discover that GPS can explain 12.4%of the difference in muscle gain effect of impedance training,and the probability of individuals with a score greater than 32 points to achieve high muscle gain effect after resistance training was 4.27 times that of low group(P<0.01).Conclusion:the GPS model constructed by 6 SNPs can be used to predict the muscle building effect of sedentary Han youth,and sedentary Han youth with GPS greater than 32 points may achieve better muscle gain effect in resistance training.
