洗手液抑菌率测试的影响因素探究

摘  要:为了探究洗手液抑菌率测试过程中不同菌种、菌株来源、测试温度、配制样品的水硬度等因素对抑菌结果的影响程度,提高抑菌率测试结果的重现性,参照GB 15979-2002《一次性卫生用品卫生规范》中的抑菌率测试方法对10款市售及自制洗手液进行了金黄色葡萄球菌和大肠杆菌的抑菌率测试,并以大肠杆菌为实验菌株探究了菌株来源、测试温度、配制样品水硬度变化时样品的抑菌率是否发生变化。结果显示,所测10款样品对金黄色葡萄球菌的抑菌率均>99.99%,但对大肠杆菌的抑菌率参差不齐。不同来源的大肠杆菌在相同测试条件下的抑菌率不尽相同。当测试温度和水硬度提高时,测试样品的抑菌率有不同程度的提升,其中测试温度的升高所导致的抑菌率升高较为明显。另外,使用玻璃试管和PBS缓冲液对金黄色葡萄球菌悬液进行计数时,发现计数结果明显偏低,采用PET材质试管或PBS中加入0.5%吐温80则能够消除这一现象。总的来说,在洗手液抑菌测试过程中标明菌种、菌株来源、测试温度、水硬度等信息,对同一个菌株进行测试时对相关测试条件加以控制,能有效地提高样品测试结果的重现性。The aims of this study were to explore the degree of influence of different bacterial species,the strain source,test temperature,water hardness and other factors on the bacteriostatic test results of liquid hand sanitizer,so as to improve the reproducibility of bacteriostatic rate test results.According to the bacteriostatic rate test method of GB 15979-2002"Hygienic standard for disposable sanitary products",the bacteriostasis of Staphylococcus aureu and Escherichia coli was tested on ten commercial and homemade liquid hand sanitizers.Escherichia coliwas used as the experimental strain to explore whether the bacteriostatic rate changed when the bacterial strain source,test temperature and the water hardness changed.The results showed that the bacteriostatic rates of the ten tested samples against Staphylococcus aureus were all above 99.99%,while the bacteriostatic rates against Escherichia coli were uneven.The bacteriostatic rates of Escherichia coli from different sources under the same test conditions were not the same.When the temperature and water hardness increased,the bacteriostatic rate of tested samples also increased in varying degrees.The bacteriostatic rate increased more obviously with the increase of test temperature.In addition,the results were significantly reduced when the Staphycolcoccus aureus suspension was counted by glass test tube and PBS buffer,which could be eliminated by using PET test tube or adding 0.5%tween 80 into PBS.In general,in the process of bacteriostatic test of liquid hand sanitizer,indicating the bacterial species,the strain source,test temperature,water hardness and other information is very important.Controlling the relevant test conditions when testing the same strain can effectively improve the reproducibility of sample test results.
