摘  要:目的 研究ε-多聚赖氨酸(EPL)/PCL/HA复合支架组织再生以及对促炎和消炎细胞浸润的影响,进而探究其体内免疫调节功能。方法 将PCL支架、PCL/HA复合支架和EPL/PCL/HA复合支架,分别移植到兔椎旁肌袋中。标本脱钙后行免疫组织化学染色及HE染色。分析T-bet,GATA3,RORγT,CD68,CCR7和CD206的水平,分别作为Th1、Th2、Th17、总巨噬细胞、I型巨噬细胞和II型巨噬细胞的标志物。结果 与PCL和PCL/HA相比,EPL/PCL/HA在体内促进组织再生,促炎性Th1和Th17浸润减少,抗炎性Th2和II型巨噬细胞浸润到再生区域,EPL/PCL/HA促进了巨噬细胞极化为M2型。结论 EPL/PCL/HA复合支架在体内具有良好的免疫调节功能及生物相容性。Objective To study the tissue regeneration of the EPL/PCL/HA composite scaffold and its immunogenic reaction. Methods The EPL/PCL/HA composite scaffold, PCL scaffold and PCL/HA composite scaffold were implanted into the rabbit paravertebral muscle bag. Samples were taken, decalcified and stained with H&E and immunohistochemistry.Immunohistochemistry(IHC) analysis was used to study the immune response of the stents. The levels of T-bet, GATA3,RORγT, CD68, CCR7 and CD206 were analyzed and used as markers of Th1, Th2, Th17, pan-macrophages, type I macrophages and type II macrophages, respectively. Results The EPL/PCL/HA scaffold inhibited the levels of proinflammatory Th1 and Th17, and promoted the infiltration of anti-inflammatory Th2 into the regeneration area. It also inhibited the pan-macrophage levels and promoted the pan-macrophage polarization into the anti-inflammatory M2 cells.Conclusions The EPL/PCL/HA composite scaffold has good biocompatibility and anti-inflammatory functions in vivo.
