摘 要:目的 探讨p120ctn-shRNA对人唾液腺腺样囊性癌细胞增殖、迁移及侵袭能力的影响,以及p120ctn-shRNA对E-钙粘蛋白和N-钙粘蛋白表达的影响。方法 选取人唾液腺腺样囊性癌细胞SACC-83细胞系进行培养,p120ctn-shRNA慢病毒表达载体转染SACC-83,同时转染空载体NC shRNA作为对照组,嘌呤霉素筛选稳定表达p120ctn-shRNA的细胞系。通过RT-PCR和Western Blot检测敲低效果。分别通过CCK8、划痕实验、迁移实验和侵袭实验观察p120连环蛋白基因敲低对人唾液腺腺样囊性癌细胞SACC-83增殖、迁移及侵袭等恶性生物学行为的影响。通过RT-PCR和Western Blot检测p120连环蛋白基因敲低对E-钙粘蛋白和N-钙粘蛋白表达的影响。结果 与对照组相比,实验组p120连环蛋白的基因表达量和蛋白表达量都大幅度降低。与对照组相比,实验组的增殖能力明显降低(P<0.05),与对照组相比,实验组的迁移能力明显降低。与对照组相比,实验组的侵袭能力明显降低(P<0.01)。E-钙粘蛋白表达略微下降,N-钙粘蛋白表达显著下降。结论 抑制p120连环蛋白的表达可以抑制人唾液腺腺样囊性癌SACC-83的增殖、迁移及侵袭的能力,同时可以较大程度下调N-cad的表达,较小程度下调E-cad的表达。Objective To investigate the effect of p120-catenin on proliferation, migration and invasion of human salivary adenoid cystic carcinoma cells. Methods Human salivary gland adenoid cystic carcinoma cell line SACC-83 was selected for culture. SACC-83 was transfected with lentiviral expression vector of p120ctn-shRNA, and the control vector NC shRNA was transfected as the control group. The cell line expressing p120ctn-shRNA was screened with purinomycin.The knockdown effect was detected by RT-PCR and Western Blot. CCK8, scratch test, migration test and invasion test were used to observe the effect of p120-catenin knockdown on the proliferation, migration and invasion of SACC-83. The effect of p120-catenin knockdown on the expression of E-cadherin and N-cadherin was detected by RT-PCR and Western Blot.Results Compared with the control group, the gene and protein expression of p120-catenin in the experimental group was significantly reduced. Compared with the control group, the expression of p120-catenin in the experimental group was down regulated. The proliferation, invasion and migration ability of the experimental group were significantly reduced(P < 0.05).E-cadherin and N-cadherin expression was decreased. Conclusions Inhibition of p120-catenin expression can inhibit the proliferation, migration and invasion of SACC-83. At the same time, it can down-regulate the expression of N-cad and E-cad.
