摘  要:目的 探索人真皮成纤维细胞在长波紫外线(UVA)辐照后的转录组学动态变化过程,为寻找皮肤光衰老模型新的分子标志物提供数据支撑。方法 体外培养原代人真皮成纤维细胞,并利用UVA分别辐照1、3和5 d后提取总RNA。制备mRNA文库并利用RNA-Seq技术测定辐照组与对照组细胞全转录组。随后计算机软件工具分析差异表达基因并进行GO功能富集分析和KEGG通路分析。结果 UVA辐照人真皮成纤维细胞1、3和5 d后,共鉴定出差异表达基因数目分别为6、668和865个,其中表达上调分别为3、50和122个,表达下调分别分别为3、618和743个;对差异基因的GO和KEGG分析表明,UVA辐照引起的基因表达差异集中于细胞外基质代谢等通路。结论 UVA辐照可引起成纤维细胞转录组学呈动态变化,未来抗衰产品研发中应深入挖掘潜在分子标志物。Objective To explore the dynamic changes in transcriptome of human dermal fibroblast after long-wave ultraviolet irradiation(UVA), thus providing support for searching for new molecular markers. Methods Primary human dermal fibroblasts were incubated in vitro and irradiated with UVA for 1, 3 and 5 days respectively, when total RNA was extracted. mRNA library was prepared and the whole transcriptomes of irradiated and control groups were sequenced by RNA-Seq. Differentially expressed genes were identified and enriched according to gene ontology(GO) terms and KEGG pathways by computer. Results After irradiation for 1, 3 and 5 days by UVA, human dermal fibroblasts were identified to have 6, 668 and 865 differentially expressed genes respectively, out of which 3, 50 and 122 genes were upregulated while 3, 618 and 743 genes were downregulated. GO function annotations and KEGG pathway analysis showed that UVA irradiation-induced differentially expressed genes were enriched in the metabolic pathways for extracellular matrix. Conclusion UVA irradiation can induce dynamic changes in the transcriptome of human dermal fibroblast. Further studies on potential molecular markers are warranted in future research and development of anti-aging products.
