膀胱癌组织miR-361-5p、BTF3表达变化及临床意义

摘　　要：目的检测膀胱癌组织中微小RNA-361-5p(miR-361-5p)、通用转录因子3(BTF3)的表达水平,并探讨其临床意义。方法选取2015年3月至2017年6月本院收治的74例膀胱癌患者为研究对象,所有患者均行根治性膀胱全切术,收集癌组织及癌旁正常组织,采用荧光定量PCR技术检测miR-361-5p、BTF3 mRNA的相对表达量,采用免疫组化法检测BTF3蛋白的表达情况,根据细胞染色强度和阳性细胞比例进行定量分析,将患者分为miR-361-5p高表达组和低表达组、BTF3高表达组和低表达组,分析miR-361-5p、BTF3表达水平与膀胱癌患者临床病理参数的关系,分析患者的3年生存情况。结果膀胱癌组织中miR-361-5p水平明显低于癌旁组织(P<0.05),BTF3 mRNA水平明显高于癌旁组织(P<0.05);膀胱癌组织中BTF3蛋白的阳性表达率为62.16%(46/74),癌旁组织中BTF3蛋白的阳性表达率为17.57%(13/74),差异有统计学意义(χ^(2)=30.694,P<0.001);miR-361-5p、BTF3表达水平与肿瘤分化程度、病理分期、淋巴结转移均有显著相关性(均P<0.05);miR-361-5p高表达组和低表达组患者的中位生存时间分别为24、16个月,高表达组患者的3年累积生存率明显高于低表达组(χ^(2)=7.516,P=0.006);BTF3高表达组和低表达组患者的中位生存时间分别为15.0、23.5个月,BTF3高表达组患者的3年累积生存率明显低于低表达组(χ^(2)=5.217,P=0.022)。结论膀胱癌组织中miR-361-5p表达降低,BTF3表达升高,其水平变化与肿瘤分期、分化程度、淋巴结转移有关,可能参与肿瘤增殖分化、转移侵袭过程,并可作为反映膀胱癌患者预后的生物指标。Objective To detect the expression levels of microRNA-361-5p(miR-361-5p)and basic transcription factor 3(BTF3)in bladder cancer tissues and discuss their clinical significance.Methods Seventy-four cases of bladder cancer patients diagnosed at our hospital from March 2015 to June 2017 as the research objects.All patients underwent radical total cystectomy to collect cancerous tissues and normal adjacent tissues.The relative expression of miR-361-5p and BTF3 mRNA was detected with fluorescence quantitative PCR technology,the expression of BTF3 protein was detected with immunohistochemical method,the relationship between the expression levels of miR-361-5p and BTF3 and clinicopathological parameters in patients with bladder cancer was analyzed,According to the cell staining intensity and the proportion of positive cells for semi-quantitative analysis,the patients were divided into miR-361-5p high expression group and miR-361-5p low expression group,BTF3 high expression group and BTF3 low expression group.the 3-year survival status of patients was analyzed.Results The level of miR-361-5p in bladder cancer tissue was significantly lower than that in adjacent tissues(P<0.05),and the level of BTF3 mRNA was significantly higher than that in adjacent tissues(P<0.05).The positive expression rate of BTF3 protein in bladder cancer tissue was 62.16%(46/74),and the positive expression rate of BTF3 protein in adjacent tissues was 17.57%(13/74),the differences were statistically significant(P<0.001).The expression levels of miR-361-5p and BTF3 were significantly correlated with tumor differentiation,pathological stage,and lymph node metastasis(P<0.05);the median survival time of miR-361-5p high-expression group and low-expression group were 24 months and 16 months,respectively,the 3-year cumulative survival rate of patients in the high-expression group was significantly higher than that in the low-expression group(P=0.006);the median survival time of patients in the BTF3 protein high expression group and low expression group was
