摘 要:目的建立肉类(猪、羊、牛肉)中20种β受体阻断剂的同位素标记-超高效液相色谱-串联质谱快速检测方法。方法样品经三氯乙酸酸解提取后,经Agilent Captiva EMR-Lipid柱净化,再经Waters BEH C18色谱柱分离,ESI源正离子模式进行多反应监测(MRM),内标法定量。结果20种β受体阻断剂线性范围为0.5~20μg/L,相关系数大于0.996,检出限为0.01~0.10μg/kg,定量限为0.03~0.30μg/kg。3个加标水平(1.0μg/kg、5.0μg/kg、10μg/kg)的回收率在80.3%~118.7%之间,相对标准偏差为3.2%~17.2%。结论该方法前处理简单、快速、准确、灵敏度高,适用于肉类食品中β受体阻断剂的定性、定量检测。Objective To establish a method for the determination of 20β-receptor blockers in meats including pork,mutton,beef by isotope labeling-ultra-high performance liquid chromatography coupled with tandem mass spectrometry.Methods The target compounds in samples were acid hydrolyzed and extracted with trichloroacetic acid solution,then purified with Agilent Captiva EMR-Lipid column and finally separated with a Waters BEH C18 column.ESI positive ion scanning multiple reaction monitoring(MRM)mode was used for detection,and internal standard method was used for quantitative analysis.Results The linear ranges of 20 beta blockers were from 0.5 to 20μg/L,with correlation coefficient(r)﹥0.996.The limits of detection were 0.01-0.10μg/kg,and the limits of quantification were 0.03-0.30μg/kg.At the spiked levels of 1μg/kg,5μg/kg,10μg/kg,the recoveries of 20 beta blockers ranged from 80.3%to 118.7%,with the relative standard deviation from 3.2%-17.2%.Conclusion The method is simple,quick,accurate and sensitive,and it is suitable for the confirmation and quantification of beta receptor blockers in meat samples.
