高分子量透明质酸抑制横纹肌肉瘤细胞迁移的初步研究

摘  要:横纹肌肉瘤是一种常见软组织肉瘤,具有恶性程度高,易转移,治疗困难等特点,严重威胁健康。采用细胞划痕试验证明高分子量透明质酸可抑制人恶性胚胎横纹肌瘤细胞(RD细胞)迁移。进一步采用荧光定量RT-PCR检测不同透明质酸受体表达,结果表明,RD细胞中CD44具有较高表达水平,且高分子量透明质酸处理可下调CD44表达。Western blot检测结果显示高分子量透明质酸可下调CD44及细胞迁移相关蛋白Cdc42和Arp2/3表达。免疫荧光超高分辨显微镜观察结果显示,CD44表达显著下调同时,RD细胞中细胞质膜边缘丝状和分枝状微丝发生解聚,表明高分子量可能通过抑制伪足形成而抑制RD细胞迁移和运动,从而为横纹肌肉瘤疾病治疗提供新思路。Rhabdomyosarcoma is a common soft tissue sarcoma, and has the characteristics of high malignancy, easy metastasis and difficult treatment, which seriously threatens the health. In this study, the cell scratch test was used to prove that high molecular weight hyaluronic acid could inhibit the migration of RD cells. Fluorescent quantitative RT-PCR was further used to detect the expression of different hyaluronic acid receptors. The results showed that CD44 had a high expression level in RD cells, and high molecular weight hyaluronic acid treatment could down-regulate the expression of CD44.The results of Western blot showed that high molecular weight hyaluronic acid could down-regulate the expression of CD44 and cell migration-related proteins Cdc42 and Arp2/3. At the same time, the results of immunofluorescence super-resolution microscopy showed that while the expression of CD44 was significantly down-regulated, the filamentous and branched microfilaments of the plasma membrane edge in RD cells depolymerized, indicating that high molecular weight might inhibit the migration and movement of RD cells by inhibiting the formation of pseudopodia, which could provide new ideas and strategies for the treatment of rhabdomyosarcoma.
