摘 要:目的 检测长链非编码(LncRNA)结肠癌相关转录物2(CCAT2)和微小RNA-200a-3p(miR-200a-3p)在宫颈癌(CC)患者血清中的表达水平,探讨其辅助诊断价值。方法 收集102例CC患者、100例健康对照者的血清标本。采用实时荧光定量聚合酶链式反应(RT-qPCR)的方法分别检测原发性CC患者、健康对照者血清LncRNA CCAT2、miR-200a-3p的表达水平,分析其与临床病理特征的关系。采用Pearson检验分析LncRNA CCAT2、miR-200a-3p在CC中表达水平的相关性。采用Kaplan-Meier生存曲线分析LncRNA CCAT2、miR-200a-3p不同表达水平对CC患者生存的影响;应用Kaplan-Meier法和Cox比例风险回归模型分别进行单因素、多因素分析影响CC患者生存的独立风险因素。结果 原发性CC患者血清中LncRNA CCAT2、miR-200a-3p的相对表达水平分别为1.757(1.023~2.314),1.957(1.125~3.471),健康对照组血清中CCAT2、miR-200a-3p的相对表达水平分别为0.766(0.531~1.032),0.965(0.667~1.495),两者比较差异有统计学意义(均P<0.001)。CCAT2的相对表达水平与CC患者肿瘤最大径、国际妇产科联合会(FIGO)分期、淋巴结是否转移、鳞状上皮细胞抗原(SCC-Ag)有关(均P<0.05),而miR-200a-3p的相对表达水平与患者FIGO分期、淋巴结是否转移、SCC-Ag有关(P均<0.05)。原发性CC患者血清CCAT2与miR-200a-3p相对表达水平呈正相关(P=0.035,r2=0.044)。CCAT2高表达水平组CC患者生存率明显低于CCAT2低表达水平组CC患者生存率,差异有统计学意义(P=0.0012),miR-200a-3p高表达水平组CC患者生存率与miR-200a-3p低表达水平组CC患者生存率差异无统计学意义(P=0.259)。单因素分析显示,淋巴结转移(P<0.001)和CCAT2(P=0.001)与总生存期(OS)相关。多因素分析显示,淋巴结转移(HR=5.568,95%CI:2.284~13.573,P<0.001)和LncRNA CCAT2高表达(HR=3.571,95%CI:1.174~10.863,P=0.025)是与OS相关的独立风险因素。结论 LncRNA CCAT2、miR-200a-3p在CC患者血清中明显升高,与CC的发生发展有一�Objective To detect the expression levels of LncRNA colon cancer related transcript 2(CCAT2)and microRNA-200a-3p(miR-200a-3p)in the serum of patients with cervical cancer and to explore their auxiliary diagnostic value.Methods Serum samples from 102 patients with primary cervical cancer and 100 healthy controls were collected.The expression levels of serum LncRNA CCAT2 and miR-200a-3p in patients with primary cervical cancer and healthy controls were detected by real-time fluorescence quantitative PCR(RT-qPCR).Pearson test was used to analyze the correlation between the expression of LncRNA CCAT2 and miR-200a-3p in cervical cancer.Kaplan Meier survival curve was used to analyze the effects of different expression levels of LncRNA CCAT2 and miR-200a-3p on the survival of cervical cancer patients;Kaplan-Meier method and Cox proportional hazards regression model were used to analyze the independent risk factors affecting the survival of cervical cancer patients.Results The relative expressions of LncRNA CCAT2 and miR-200a-3p in serum of patients with primary cervical cancer were 1.757(1.023,2.314)and 1.957(1.125,3.471),respectively.The relative expressions of LncRNA CCAT2 and miR-200a-3p in serum of healthy control group were 0.766(0.531,1.032)and 0.965(0.667,1.495),respectively.There was significant difference between the two groups(all P<0.001).The relative expression of LncRNA CCAT2 was related to tumor size,FIGO stage,lymph node metastasis and Squamous Cell Carcinoma Antigen(SCC Ag)(all P<0.05),while the relative expression of miR-200a-3p was related to FIGO stage,lymph node metastasis and SCC Ag(all P<0.05).There was a positive correlation between the relative expression of serum LncRNA CCAT2 and miR-200a-3p in patients with primary CC(P=0.035,r 2=0.044).The survival rate of CC patients in LncRNA CCAT2 high expression group was significantly lower than that in LncRNA CCAT2 low expression group(P=0.0012).There was no significant difference between the survival rate of cervical cancer patients in miR-200a-3p high
