摘　　要：目的:分析肝细胞癌(HCC)中Apaf-1相互作用蛋白(APIP)的表达情况,并分析其与HCC患者临床病理特征及预后的相关性。方法:分别于肿瘤基因组图谱(TCGA)、基因型组织表达数据库(GTEx)、基因表达综合(GEO)数据库中收集APIP在正常肝组织、肝细胞癌组织中的表达,对2315例非癌样本和3508例癌样本进行分析。应用实时荧光定量聚合酶链反应(RTqPCR)检测HCC细胞株(SK-HEP-1、Huh7)和正常肝细胞株(L02)中APIP mRNA表达,免疫组织化学实验检测91例HCC组织及82例癌旁肝组织中APIP蛋白表达水平。并使用GEPIA数据库对APIP进行生存分析。同时,基于TCGA数据库的表达数据及相应临床病理参数构建单因素、多因素Cox回归及列线图预测模型,评估HCC潜在危险因素及患者预后。结果:公共数据整合分析结果显示,APIP在HCC组织中表达上调[SMD=0.87(0.64~1.11),I2=91.2%,P<0.001]。RT-qPCR结果显示,与正常肝细胞株相比,APIP mRNA在HCC细胞株中呈高表达(P<0.001);免疫组织化学实验结果显示,APIP蛋白在HCC组织中表达水平高于癌旁肝组织(P<0.001)。生存分析及列线图预测模型均提示APIP高表达与HCC患者不良预后相关。结论:APIP在HCC中呈高表达,且与HCC患者的不良预后相关,可能是HCC的诊断和预后的标记物。Objective:To analyze the expression of the Apaf-1 interacting protein(APIP)in Hepatocellular carcinoma(HCC)and its correlation with clinicopathological features and prognosis of HCC patients.Methods:The expression values of APIP in normal liver tissues and hepatocellular carcinoma tissues were collected respectively from Tumor Genome Atlas(TCGA),Genotype Tissue Expression(GTEx)database,and Gene Expression Synthesis(GEO)databases.2,315 non-cancer samples and 3,508 cancer samples were analyzed.Real-time quantitative polymerase chain reaction(RT-qPCR)was used to detect the mRNA level of APIP in HCC cell lines(SKHEP-1,Huh7)and normal liver cell lines(L02).Immunohistochemistry(IHC)assay was used to detect the protein level of APIP in 91 HCC tissues and 82 adjacent liver tissues to verify the expression of APIP in HCC.Survival analysis of APIP was performed using GEPIA database.At the same time,univariate,multivariate Cox regression and nomogram prediction models were constructed based on the expression data of TCGA database and corresponding clinicopathological parameters to evaluate the potential risk factors and prognosis of HCC patients.Results:The results of public data integration analysis showed that APIP expression was up-regulated in HCC tissues[SMD=0.87(0.64-1.11),I2=91.2%,P<0.001].RT-qPCR results showed that APIP mRNA was highly expressed in HCC cell lines compared with normal liver cell lines(P<0.001).Immunohistochemical results showed that the expression level of APIP protein in HCC tissues was higher than that in paracancer liver tissues(P<0.001).Both survival analysis and nomogram prediction model indicated that high expression of APIP was associated with poor prognosis in HCC patients.Conclusion:APIP is highly expressed in HCC.It is correlated with poor prognosis of HCC patients and may be diagnostic and prognostic marker for HCC.
