摘 要:选取14份市售鳕鱼肉松样品,采用Tiangen试剂盒、Takara试剂盒和十二烷基磺酸钠(SDS)3种提取方法对样品中DNA进行提取,对比分析不同提取方法对DNA纯度、得率及PCR扩增效果的影响。结果表明,Takara试剂盒法所得的DNA纯度较低,SDS法的PCR扩增效果低于Tiangen试剂盒法;Tiangen试剂盒法的DNA得率最高,且A260/A280为1.80,A260/A230为1.10,对荧光定量PCR影响最小,满足鳕鱼制品鉴定需要。因此,Tiangen试剂盒法是一种较好的DNA提取方法。14 commercially available codfish pine samples were selected and three DNA extraction methods(Tiangen kit,Takara kit,SDS)were applied to extract DNA.The DNA purity,yield and PCR amplification effect of different extraction methods were evaluated.The results showed that the DNA purity of Takara kit was low,the PCR amplification effect of SDS was lower than the Tiangen kit;the DNA extraction rate of Tiangen kit was the highest,and the value of A260/A280and A260/A230were 1.80 and 1.10,respectively,which had the least impact on quantitative PCR and met the requirements of codfish products.Therefore,Tiangen kit method was a better method for DNA extraction.
