摘 要:为了最大限度提升毛叶木姜子茎段腋芽萌发有效芽的诱导率,降低褐变对有效芽的影响,本研究采用单因素试验和Box-Behnken响应面法,探究不同浓度的6-苄基氨基嘌呤(6-BA)、苯基脲类衍生物(TDZ)和聚乙烯比咯烷酮(PVP)对茎段腋芽诱导褐化率和有效芽诱导率的影响。结果表明,3个因素对腋芽诱导褐化率的降低程度为PVP>TDZ>6-BA,对提升有效芽诱导率的促进程度为6-BA>TDZ>PVP;最佳的茎段腋芽诱导培养条件为B5+6-BA 1.80 mg/L+TDZ 0.04 mg/L+PVP 1.30 g/L,最优有效芽诱导率为79.39%,褐化率为15.71%。实际验证结果有效芽诱导率为77.78%,褐化率为17.78%。本研究结果不仅为毛叶木姜子茎段腋芽诱导培养条件提供了最优路径,也为后续培养条件的优化拓宽了可靠的技术手段。In order to maximize the induction rate of effective buds in axillary bud germination of stem segments of Litsea mollis Hemsl.and reduce the effect of browning on effective buds,we used single factor test and Box Behnken response surface method to explore the effects of different concentrations of 6-Benzylaminopurine(6-BA),phenylurea derivatives(TDZ)and polyvinylpyrrolidone(PVP)on the browning rate and effective bud induction rate of axillary buds.The results showed that the three factors reducing the browning rate of axillary buds was PVP>TDZ>6-BA,and promoting the effective bud induction rate was 6-BA>TDZ>PVP.The optimum conditions for inducing axillary buds in stem segments were B5+6+BA 1.80 mg/L+TDZ 0.04 mg/L+PVP 1.30 g/L.The optimum effective bud induction rate was 79.39%and the browning rate was 15.71%.The verification results showed that the effective bud induction rate was 77.78%and the browning rate was 17.78%.The results of this study only provided the optimal path for the induction and culture conditions of axillary buds in stem segments of Litsea mollis Hemsl.,but also broadened the reliable technical means for the optimization of subsequent culture conditions.
