摘 要:目的筛选肺腺癌中差异表达的环状非编码RNA(miRNA),并分析其对肺腺癌(LUAD)迁移的影响及潜在机制。方法通过miRNA芯片筛选出LUAD组织和正常肺组织中差异表达的miRNA并预测其潜在的生物学功能和信号通路;通过荧光定量逆转录聚合酶链反应(qRT-PCR)和癌症基因组图谱(TCGA)分析验证hsa-let-7e-5p在LUAD组织和细胞系中的表达水平;通过划痕试验检测hsa-let-7e-5p对LUAD细胞迁移的影响;通过生物信息学分析和qRT-PCR验证hsa-let-7e-5p与基因DTX2、NME6、C8orf58、GATM和DHX57之间的靶向关系。结果miRNA芯片结果显示肺腺癌组织中347个miRNA下调而229个miRNA上调。hsa-let-7e-5p在LUAD组织和细胞系中下调,过表达hsa-let-7e-5p可以抑制肺腺癌细胞迁移。结论hsa-let-7e-5p在肺腺癌中低表达并抑制肺腺癌细胞的迁移,DTX2、NME6、C8orf58、GATM和DHX57是hsa-let-7e-5p的潜在靶基因。Objective To screen differentially expressed miRNAs and explore its effect and mechanism on cell migration in lung adenocarcinoma(LUAD).Methods Differentially expressed miRNAs in LUAD tissues and normal lung tissues were screened by miRNA microarrays,and then bioinformatics analysis was used to predict their potential biological functions and signaling pathways.The cancer genome atlas(TCGA)analysis and quantitative real-time PCR(qRT-PCR)verified the expression level of hsa-let-7 e-5 p in LUAD tissues and cell lines.The effect of hsa-let-7 e-5 p cell migration in LUAD was examined by would healing experiment.After screening the underlying target genes by bioinformatics analysis,the targeting relationship between hsa-let-7 e-5 p and DTX2,NME6,C8 orf58,GATM and DHX57 were verified by qRT-PCR.Results The miRNA microarray results showed that 347 miRNAs were down-regulated while 229 miRNAs were up-regulated in lung adenocarcinoma tissues.Compared with normal lung tissue and cells,the expression level of hsa-let-7 e-5 p was significantly down-regulated.Besides,overexpression of hsa-let-7 e-5 p inhibitedLUAD cell migration.Conclusion Non-coding RNA hsa-let-7 e-5 p is down-regulated in LUAD and inhibits the migration of lung adenocarcinoma cells.DTX2,NME6,C8 orf58,GATM and DHX57 are the potential target genes of hsa-let-7 e-5 p.
