摘 要:目的:探讨骨髓间充质干细胞(Bone Marrow Mesenchymal Stem Cells,MSCs)对淋巴内皮祖细胞(Lymphatic Endothelial Progenitor Cells,LEPCs)迁移能力的影响。方法:选择C57BL/6小鼠股骨和胫骨提取MSCs和LEPCs,贴壁培养法细胞进行培养及传代。流式细胞仪分选MSCs与LEPCs的细胞表型,获得目标细胞,并进行分化功能鉴定。通过划痕实验与Transwell迁移实验检测MSCs对LEPCs细胞迁移作用。结果:通过贴壁培养法,从C57BL/6小鼠股骨和胫骨能够成功提取出足够数量的MSCs和LEPCs。MSCs与LEPCs表型分别为Sca-1、CD29阳性,CD45、CD11b为阴性和CD34、CD133、VEGFR-3均阳性。MSCs可分化为成骨细胞、软骨和脂肪细胞。Transwell迁移实验与细胞划痕实验示,MSCs与LEPCs共培养细胞的迁移率实验组高于对照组(P<0.05)。结论:MSCs与LEPCs可从小鼠骨髓中分离,且易于体外培养和传代;MSCs可以促进LEPCs的迁移能力。Objective:To explore the effect of bone marrow mesenchymal stem cells on the migration ability of lymphatic endothelial progenitor cells.Methods:MSCs and LEPCs were extracted from femur and tibia of C57BL/6 mice,and the cells were cultured and subcultured by adherent culture.The phenotypes of MSCs and LEPCs were sorted by flow cytometry,and the target cells were obtained,and the differentiation function was identified.The effect of MSCs on LEPCs fine cell migration was detected by wounding healing and Transwell migration test.Results:Sufficient amounts of MSCs and LEPCs were successfully extracted from femur and tibia of C57BL/6 mice by adherent culture.The phenotypes of MSCs and LEPCs were positive for Sca-1 and CD29,negative for CD45 and CD11b and positive for CD34,CD133 and VEGFR-3,respectively.MSCs can differentiate into osteoblasts,cartilage and adipocytes.Transwell migration test and wounding healing showed that the migration rate of MSCs and LEPCs co-cultured cells in the experimental group was significantly higher than that in the control group(P<0.05).Conclusion:MSCs and LEPCs can be isolated from mouse bone marrow and are easy to be cultured and passaged in vitro.MSCs can promote the migration ability of LEPCs.
