摘  要:目的:研究雪菊茶总黄酮对家兔离体十二指肠收缩的抑制作用及机制。方法:采用Medlab生物信号系统和分光光度计检测雪菊茶总黄酮对离体兔肠收缩抑制及机制。结果:雪菊茶总黄酮能抑制兔离体肠收缩。20 mg/mL的雪菊茶总黄酮能协同(P<0.05)AD抑制肠收缩,能显著(P<0.05)抑制CaCl_(2)对其促进作用。紫外-可见分光光度检测显示,雪菊茶总黄酮3个浓度组家兔离体肠中ChAT活性均低于正常对照组,A-CHE和TNOS活性均升高。结论:黄酮是雪菊茶中抑制十二指肠收缩的有效成分,作用机制与激动β受体、促进NO合成从而激活GC-cGMP-PKG信号通路、抑制ACh从而抑制AC-cAMP-PKA信号通路、抑制肌膜I_(Ca-L)从而抑制胞外Ca^(2+)内流等有关。Objective:To investigate the effects of the flavonoids of Coreopsis tinctoria Nutt.(CTFS)on the contractile activity of the rabbit duodenum in vitro,and to explore the mechanism.Methods:The Medlab biosignal system and spectrophotometer were used to detect the inhibition and mechanism of CTFS on contractile activity of the rabbit duodenum in vitro.Results:The CTFS could inhibit the contraction of duodenum.The CTFS could synergize(P<0.05)the inhibition effect of AD on the intestinal contraction,and significantly(P<0.05)inhibit the promotion effect of Ca Cl_(2).The UV-vis detection showed that the Ch AT activity of the three concentration groups of CTFS was decreased,the activities of A-CHE and TNOS were increased,compared with the normal control group,.Conclusion:The fiavonoids were effective component of Coreopsis tinctoria in inhibiting the duodenum movement.The mechanism may be related to the inhibitions of Ach,AC-c AMP-PKA signal pathway and I_(Ca-L) in sarcolemma,the promotion of NO synthesis and the activations of GC-c GMP-PKG signaling pathway andβreceptor.
