摘  要:对箬竹叶多糖进行DEAE-52离子层析纯化,并研究其多糖的理化特性以及生物活性。箬竹叶多糖纯化后得到中性多糖和酸性多糖,中性多糖含有少量的蛋白质,不含糖醛酸、硫酸基和氨基糖,其重均分子量在5.0×10^(4)~1.5×10^(5) Da之间,酸性多糖含有蛋白质、糖醛酸和硫酸基,不含氨基糖,其重均分子量在2.3×10^(4)~1.8×10^(5) Da之间。中性多糖是由岩藻糖、木糖、甘露糖、葡萄糖和半乳糖构成,其物质量比为1.4∶1.6∶0.6∶1.7∶1.0;酸性多糖由岩藻糖、木糖、甘露糖、葡萄糖、半乳糖和葡萄糖醛酸及半乳糖醛酸构成,其物质量比为1.3∶1.5∶0.8∶1.5∶1.3∶1.8∶0.6。在体内抗氧化试验和体外抗氧化试验中,酸性多糖具有更强的抗氧能力,其可以提高T-AOC、SOD及GSH-px含量,显著降低MDA含量;可以较好地清除·OH和DPPH自由基。降血脂试验中表明,酸性多糖对肝胆盐的结合能力更好。因此酸性多糖具有较好的生物学活性,可以作为天然的功能性食品添加剂。The polysaccharide from Indocalamus leaves was purified by DEAE-52 ion chromatography,and its physicochemical properties and biological activities were studied.The neutral and acidic polysaccharide were obtained after the purification.The neutral polysaccharide contained a little amount of protein,no uronic acid,sulfate or amino sugar,and its weight average molecular weight was 5.0×10^(4)~1.5×10^(5) Da,the acidic polysaccharide contained a little amount of protein,uronic acid and sulfate group,contained no amino sugar,and its weight average molecular weight was 2.3×10^(4)~1.8×10^(5) Da.Neutral polysaccharide was composed of fucose,xylose,mannose,glucose and galactose,and its mass ratio was 1.4∶1.6∶0.6∶1.7∶1.0;The acidic polysaccharide was composed of fucose,xylose,mannose,glucose,galactose,glucuronic acid and galacturonic acid,and its mass ratio was 1.3∶1.5∶0.8∶1.5∶1.3∶1.8∶0.6.In antioxidant test in vivo and vitro,acidic polysaccharide had stronger antioxidant ability,which could significantly increase the contents of T-AOC,SOD and GSH-px and reduce the content of MDA,and had strong free radical scavenging ability of·OH and DPPH.The hypolipidemic test showed that acidic polysaccharide had better binding ability to hepatobiliary salt.Therefore,acidic polysaccharide had good biological activities and could be used as a natural functional food additive.
