TCDD诱导的腭裂小鼠腭突Wnt5a与IL-10的表达变化研究

摘  要:目的研究Wnt5a与白细胞介素(interleukin,IL)-10的表达在2,3,7,8-四氯二苯并二噁英(2,3,7,8-tetrachlorodibenzo-p-dioxin,TCDD)致胎鼠腭裂中的作用。方法24只C57BL/6J怀孕小鼠随机分为TCDD实验组和玉米油对照组,每组12只。在胚胎形成的10.5天(E10.5),TCDD组通过口饲灌胃方式一次性给予孕鼠64μg/kg的TCDD(溶于总量为0.2 mL的玉米油中),对照组通过口饲灌胃方式给予等量玉米油。分别获取E13.5,E14.5和E15.5的胎鼠腭突组织,石蜡切片,HE染色,光镜观察腭突组织融合情况;Western-blot检测腭突组织中Wnt5a和IL-10的蛋白表达。结果HE染色观察到腭突组织未融合则证明腭裂小鼠建模成功。在E13.5天,E14.5天和E15.5天,与对照组相比TCDD组胎鼠出现腭突发育迟缓,腭裂形成。在各时间点,TCDD组Wnt5a相对表达量较对照组显著减少[(0.37±0.05)比(0.47±0.07),(0.32±0.07)比(0.39±0.05),(0.27±0.06)比(0.33±0.04),P值分别为0.005,0.042,0.046];IL-10相对表达量较对照组显著增加[(0.24±0.02)比(0.15±0.01);(0.29±0.03)比(0.25±0.04);(0.90±0.20)比(0.54±0.12),P值分别为0.000,0.043,0.011],且随着时间推移Wnt5a和IL-10呈现出明显的时序性减少(F=4.65,P=0.025)和增加(F=53.01,P=0.000)。结论Wnt5a和IL-10在TCDD诱导胎鼠腭裂的形成过程中具有重要作用。Objective To study the role of Wnt5a and interleukin(IL)-10 expression in cleft palate induced by 2,3,7,8-tetrachlorodibenzo dioxin(TCDD)in fetal mice.Methods Twenty-four pregnant C57BL/6J mice were randomly divided into TCDD experimental and corn oil control groups with twelve mice in each gourp.Pregnant mice in TCDD group was given 64μg/kg TCDD(dissolved in 0.2 mL corn oil)by oral gavage on 10.5 day after embryo formation(E10.5),the control group was given the same amount of corn oil.The fetal mice palatal process tissues of E13.5,E14.5 and e15.5 were obtained respectively.Paraffin section,HE staining and light microscope were used to observe the fusion of palatal process.The protein expressions of Wnt5a and IL-10 in the palatal process of fetal mice were detected by Western blot.Results HE staining showed that the palatal tissue was not fused,which proved that the model of cleft palate mice was successful.At E13.5,E14.5 and E15.5,compared with the control group,fetal mice in TCDD group showed palatal retardation and cleft palate formation.At each time point,compared with the control group,the relative expression of Wnt5a in TCDD group was significantly decreased[(0.24±0.02)vs(0.15±0.01),(0.29±0.03)vs(0.25±0.04),(0.90±0.20)vs(0.54±0.12),P values were 0.000,0.043,0.011,respectively].Compared with the control group,the relative expression of IL-10 in TCDD group was significantly increased[(0.24±0.02)vs(0.15±0.01),(0.29±0.03)vs(0.25±0.04),(0.90±0.20)vs(0.54±0.12),P values were 0.000,0.043,0.011,respectively].And by the passage of time E13.5,E14.5,E15.5,there was a significant sequential decrease(F=4.65,P=0.025)with the expression of Wnt5a protein and increase(F=53.01,P=0.000)with the expression of IL-10 protein in TCDD group.Conclusion Wnt5a and IL-10 may play an important role in the formation of fetal mice cleft palate induced by TCDD.
