摘　　要：目的 探索组织金属蛋白酶抑制剂1(TIMP1)基因表达与结直肠癌预后及免疫浸润的相关性。方法 使用癌症基因组图谱(TCGA)数据库RNA表达数据分析TIMP1基因表达水平与结直肠癌患者临床病理特征的相关性,并分析TIMP1在结直肠癌和正常组织中的表达。采用Kaplan-Meier生存分析和Cox回归分析TIMP1与预后的相关性。采用基因集合富集分析(GSEA)和单样本基因集富集分析(ssGSEA)研究TIMP1的富集通路和功能,并进行免疫细胞浸润的关联分析。结果 TIMP1在结直肠癌中高表达,并和结直肠癌患者的预后不良有关,ECM通路、MAPK通路及免疫相关通路等可能是TIMP1调控的关键通路。同时,TIMP1在结直肠癌中的表达与免疫浸润水平有关。结论 TIMP1可作为结直肠癌的潜在治疗靶点和预后生物标志物。Objective To explore the correlation between TIMP1 gene expression and prognosis and immune invasion of colorectal cancer.Methods RNA expression data from the Cancer Genome Atlas(TCGA) database were used to analyze the correlation between the expression level of TIMP1 gene and clinicopathologic features of colorectal cancer patients,and to analyze the expression of TIMP1 in colorectal cancer tumors and normal tissues.Kaplan-Meier survival analysis and Cox regression were used to analyze the correlation between TIMP1 and prognosis.Gene set enrichment analysis(GSEA) and single sample gene set enrichment analysis(ss GSEA) were used to study the enrichment pathway and function of TIMP1,and association analysis of immune cell infiltration was performed.Results TIMP1 was highly expressed in colorectal cancer and was associated with poor prognosis in patients with colorectal cancer.ECM pathway,MAPK pathway and immune-related pathway might be key pathways for TIMP1 regulation.Meanwhile,the expression of TIMP1 in colorectal cancer was related to the level of immune invasion.Conclusion TIMP1 can be a potential therapeutic target and prognostic biomarker for colorectal cancer.
