摘 要:目的 探讨NLRC3(NLR family CARD domain containing 3)低表达后对人肺正常上皮细胞株BEAS-2b侵袭和迁移的影响及其机制。方法 采用Lipofectamin 2000转染试剂将NLRC3基因的小干扰片段siNLRC3转染至BEAS-2b细胞;荧光定量PCR检测干扰片段siNLRC3的干扰效率;Transwell迁移实验和Matrigel侵袭实验检测NLRC3低表达后BEAS-2b细胞迁移和侵袭能力的变化;Western Blot检测NLRC3低表达后基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)、钙粘蛋白E-cadherin表达的影响。结果 荧光定量PCR实验结果表明siNLRC3干扰效率为77%。Transwell迁移实验和Matrigel侵袭实验结果表明NLRC3低表达后BEAS-2b细胞迁移和侵袭能力增加(P<0.05)。Western blot结果显示NLRC3低表达后BEAS-2b细胞中MMP-2、MMP-9蛋白表达水平上调,E-cadherin蛋白表达水平明显下降(P<0.01)。结论 NLRC3基因低表达后可能通过降低E-cadherin蛋白表达以及促进MMP-2、MMP-9蛋白表达来增强BEAS-2b细胞的侵袭和转移能力。Objective To investigate the effect and mechanism of low expression of NLRC3(NLR family card domain containing 3) on invasion and migration of human normal lung epithelial cell line BEAS-2b.Methods The small interfering RNA of the NLRC3 gene(siNLRC3) was transfected into BEAS-2b cells using Lipofectamin 2000 transfection reagent.The interference efficiency of siNLRC3 was detected by RT-qPCR.The migration and invasion of BEAS-2b cells after low expression of NLRC3 was detected by Transwell.Western Blot was used to detect the expression of matrix metalloproteinase-2,matrix metalloproteinase-9(MMP-2,MMP-9) and E-cadherin after the expression of NLRC3 was decreased.Results The results of RT-qPCR showed that the interference efficiency of siNLRC3 was 77%.Transwell assay showed that the migration and invasion ability of BEAS-2b cells increased after low expression of NLRC3(P<0.05).Western blot analysis showed that the expression of MMP-2 and MMP-9 in BEAS-2b cells was up-regulated and the expression of E-cadherin was significantly decreased(P<0.01) after low expression of NLRC3.Conclusions The low expression of NLRC3 may enhance the invasion and metastasis of BEAS-2B cells by reducing the expression of E-cadherin and promoting the expression of MMP-2 and MMP-9.
