摘 要:目的初步探寻玉女煎治疗糖尿病的作用机制。方法通过中药系统药理学分析平台(TCMSP)和中医药研究综合数据库(TCMID)检索玉女煎的化学成分和作用靶点,借助Genecards数据库、TTD数据库和DrugBank数据库预测糖尿病对应靶点,应用Cytoscape 3.7.2软件构建中药-成分-靶点-疾病网络图,借助DAVID数据库进行GO功能分析和KEGG通路分析,并将主要活性成分与抗糖尿病药物关键靶点进行分子对接。结果玉女煎治疗糖尿病化学成分257个,对应靶点982个,将其与495个疾病靶点交集得到得到134个共同靶点,其中核心靶点为11β-羟基类固醇脱氢酶1(11β-hydroxy steroid dehydrogenase type 1,11β-HSD1)、非受体型蛋白酪氨酸磷酸酶1(Recombinant Protein Tyrosine 1,PTPN1)、腺昔A1受体(Adenosine A1 Receptor,ADORA1)、性激素结合球蛋白(sex hormone-binding globulin,SHBG);分子对接显示活性成分与核心靶点结合性较好。结论玉女煎活性成分异装契皂忒(smilagenin)、菠莫皂昔元(sarsasapogenin)可能作用于11β-HSD1、ADORA1、SHBG等靶点调节多条信号通路HIF-1、VEGF、TLR等发挥抗糖尿病的作用。Objective To explore the mechanism of Yunv Decoction in treating diabetes.Methods The chemical components and action targets of Yunu decoction were searched by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and Traditional Chinese Medicines Integrated Database(TCMID),the corresponding targets of diabetes were predicted by Genecards,TTD and DrugBank,the network map of traditional Chinese medicine-components-target-disease was constructed by Cytoscape 3.7.2,the GO function and KEGG pathway were analyzed by David Database,and the main active components were matched with the key targets of Anti-diabetic medication.Results There were 982 targets corresponding to 257 chemical components of Yunv Decoction for treating diabetes mellitus,and 134 common targets were obtained by intersection of them with 495 disease targets,among which the core targets were 11β-HSD1,PTPN1,ADORA1 and SHBG.Molecular docking showed that the active ingredient had good binding ability to the core target.Conclusion The active ingredients smilagenin and Sarsasapogenin of Yunv Decoction may act on 11β-HSD1,ADORA1,SHBG and other targets to regulate multiple signaling pathways such as HIF-1,VEGF,TLR and play an anti-diabetic role.
