摘　　要：目的:探究整合素α5(integrinα5,ITGA5)在脑胶质瘤组织中的表达及其与临床病理参数和预后的关系。方法:比较TCGA数据库中胶质瘤和正常脑组织ITGA5的表达差异和临床预后的生物学信息。收集临沂市人民医院54例脑胶质瘤手术患者的脑胶质瘤组织标本和临床资料,以及20例脑外伤患者颅内减压后的脑组织标本。以外伤后的脑组织为对照,采用免疫组织化学染色法与蛋白质印迹法检测ITGA5蛋白在胶质瘤中的表达。采用统计学方法研究ITGA5表达水平与临床病理参数的相关性。通过绘制生存曲线,比较ITGA5高表达和低表达患者的预后差异。结果:TCGA数据库中ITGA5在LGG与GBM中的表达均有显著差异(P<0.001),表达量随着胶质瘤WHO病理分级的升高而增加,且与患者的不良预后相关。ITGA5在胶质瘤组织中的高表达率为59.2%(32/54),外伤后脑组织中的高表达率为5.0%(1/20),两者之间的差异有统计学意义(P<0.05)。ITGA5在胶质瘤组织中的表达与WHO分级、Ki-67和p53基因型相关(P<0.05),而与年龄、性别和肿瘤部位无关(P>0.05)。ITGA5高表达胶质瘤患者的生存时间显著低于ITGA5低表达胶质瘤患者(P<0.05)。结论:ITGA5在胶质瘤组织中的表达增加,且ITGA5的表达与胶质瘤病理分级和患者生存密切相关。Objective:To investigate the expression of integrinα5(ITGA5)in glioma tissues and its relationship with clinicopathological parameters and prognosis.Methods:Comparing the differences in the expression of ITGA5 and the biological information of clinical prognosis in glioma and normal brain tissues in the TCGA database.Collec⁃ting the glioma tissue specimens and clinical data of 54 patients with glioma surgery in Linyi People's Hospital,and the brain tissue specimens of 20 patients with traumatic brain injury after intracranial decompression.The brain tissue af⁃ter trauma was used as the control,and the expression of ITGA5 protein in glioma was detected by immunohistochemi⁃cal staining and western blotting.Statistical methods were used to study the correlation between ITGA5 expression lev⁃el and clinicopathological parameters.By drawing survival curves,the difference in prognosis of patients with high and low expression of ITGA5 was compared.Results:In the TCGA database,the expression of ITGA5 in LGG and GBM was significantly different(P<0.001),and the expression increased with the increase of the WHO pathological grade of glioma,and was related to the poor prognosis of patients.The high expression rate of ITGA5 in glioma tissue was 59.2%(32/54),and the high expression rate in brain tissue after trauma was 5.0%(1/20),and the difference was statistically significant(P<0.05).The expression of ITGA5 in glioma tissue was correlated with WHO grade,Ki-67 and p53 genotype(P<0.05),but not with age,gender and tumor site(P>0.05).The survival time of patients with gliomas with high ITGA5 expression was significantly lower than that of patients with gliomas with low ITGA5 expres⁃sion(P<0.05).Conclusion:The expression of ITGA5 is increased in glioma tissue,and the expression of ITGA5 is closely related to the pathological grade of glioma and the survival of patients.
