摘 要:为比较来源于猪骨髓来源和外周血单核细胞来源的两种树突状细胞的生物学特性,本研究分别从猪骨髓和外周血中分离前体细胞,经重组猪粒细胞集落刺激因子(GM-CSF)和重组猪白细胞介素-4(IL-4)诱导分化为猪骨髓来源树突状细胞(BMDC)和猪单核细胞衍生树突状细胞(MoDC),对这两种来源的树突状细胞进行形态学观察,细胞表型鉴定以及功能检测,并进行比较。结果显示:BMDC和MoDC均具有典型的树突状细胞形态和基本功能;BMDC和MoDC的分子表型为CD172a^(+)MHC II^(+),经LPS刺激后表达升高;BMDC和Mo DC均具有吞噬功能,BMDC吞噬能力较强;在TranswellTM迁移实验中,Mo DC具有更好的迁移能力;BMDC和MoDC均可刺激淋巴细胞增殖,BMDC刺激淋巴细胞增殖率高于MoDC。本研究可为不同体外模型中DCs的选择提供依据。To compare the biological characteristics of dendritic cells(DCs)derived from porcine bone marrow and peripheral blood mononuclear cells,precursor cells were isolated and induced to differentiate into bone marrow-derived dendritic cells(BMDC)and monocyte-derived DC(MoDC)in vitro by using recombinant-porcine granulocyte macrophage colony-stimulating factor(GM-CSF)and porcine interleukin-4(IL-4).These two sources of DCs were observed and compared for their morphology,phenotypes and functions.Both BMDC and MoDC were developed to their typical dendritic morphology and basic functions.The molecular phenotypes of both DCs were CD172a^(+)MHCⅡ^(+).Moreover,the ratios of these cell markers were obviously increased when treated with LPS.The phagocytic ability of BMDC was higher than MoDC.MoDC showed excellent migration ability in TranswellTM assay.Additionally,both DCs possessed potent T-cell stimulatory capacity,and the stimulation index of BMDC was higher than MoDC.The results from this study provided a basis for the in vitro selection of DCs in different models.
