Optimization of Basic Medium for Clostridium butyricum Rapid Proliferation by Response Surface Methodology

摘  要:丁酸梭菌是一种专性厌氧菌,具有极强的整肠作用,在食品、医药、发酵工业等多个领域中有着十分广阔的应用前景。但是由于培养条件苛刻,培养物活菌数量少,其多种应用被限制。通过单因素实验研究了5种碳源、5种氮源、4种除氧剂以及培养温度对丁酸梭菌生长的影响,并在最陡爬坡实验的基础上使用Design Expert 10.0软件研究各因素对丁酸梭菌OD值的响应面分析,最终确定最优培养基。结果表明,优化后培养基中最佳碳源为葡萄糖(14.0 g/L),最佳氮源为酵母膏(16.0 g/L),最佳除氧剂为半胱氨酸盐酸盐(0.9 g/L),最适培养温度为37℃。此时丁酸梭菌生长最快速,且最大OD600值达到3.350 4,比优化前提高约132.72%。Clostridium butyricum is a kind of obligate anaerobe, and has a extremely strong regulating intestinal function. It has a very broad application prospect in many fields, such as food, medicine, fermentation industry and so on. However, due to the harsh culture conditions and the small number of viable bacteria in the culture, its various applications are limited. The effects of five carbon sources, five nitrogen sources, four deoxidizers and culture temperature on the growth of Clostridium butyricum are studied through single factor experiment. On the basis of the steepest climbing experiment, the response surface analysis of various factors on the OD value of Clostridium butyricum is studied by using Design Expert 10.0 software, and the optimal culture medium is finally determined. The results show that the best carbon source in the optimized medium is glucose(14.0 g/L), the best nitrogen source is yeast extract(16.0 g/L), the best deoxidizer is cysteine hydrochloride(0.9 g/L), and the optimum culture temperature is 37 ℃. At this time, Clostridium butyricum grows the fastest, and the maximum OD600value reaches 3.350 4, which is about 132.72% higher than that before optimization.
