摘 要:目的利用生物信息学数据库探究NUAK2基因在乳腺癌诊疗中的临床价值并分析其对乳腺癌细胞MCF-7生物学功能的影响。方法通过TCGA和GTEx数据库分析NUAK2基因在乳腺癌中的表达水平及其与乳腺癌患者临床病理特征的相关性;以人乳腺癌细胞MCF-7为细胞模型,合成NUAK2的特异性siRNA(si-NUAK2)并转染MCF-7细胞,通过qRT-PCR和Western blot检测NUAK2的表达情况;干扰人乳腺癌MCF-7细胞NUAK2的表达,利用平板克隆实验和CCK-8实验检测细胞增殖水平,Transwell实验和划痕实验检测细胞迁移能力;通过String数据库得到NUAK2相互结合蛋白并进行功能富集分析。结果TCGA数据库和GETx数据库分析结果及患者组织标本检测结果显示NUAK2在乳腺癌中高表达且与患者临床病理分型密切相关;平板克隆实验和CCK-8实验结果显示si-NUAK2组细胞增殖能力显著低于对照组;Transwell实验和划痕实验结果显示si-NUAK2组细胞的迁移能力明显低于对照组;String数据库分析结果显示NUAK2与蛋白水解、生理节律以及Wnt等信号通路密切相关。结论NUAK2可能与乳腺癌的发生发展密切相关,有望成为乳腺癌治疗的新靶点。Objective To investigate the clinical significance of NUAK2 and its clinical features by using bioinformatics database and the effects of NUAK2 on the biological function of breast cancer MCF-7 cells.Methods The mRNA levels of NUAK2 and its clinical features were analyzed by TCGA database and GETx database.Human breast cancer MCF-7 cells were cultured and transfected with siRNA.The knockdown effect of NUAK2 in MCF-7 cells was verified by real-time fluorescence quantitative polymerase linked reaction(qRT-PCR)and Western blotting.The effects of NUAK2 on MCF-7 proliferation were verified by plate clone formation assay and CCK-8 assay.Transwell assay and scratch assay verified the effect of NUAK2 on the migration of MCF-7 in breast cancer cells.The interaction and functional enrichment of NUAK2 and its related proteins were analyzed by String database.Results The results of bioinformatics analysis and Western blotting showed that NUAK2 was highly expressed in breast cancer and closely related to clinicopathological classification.The results of qRT-PCR and Western blotting showed that the expression of NUAK2 in knockout group was significantly lower than that in control group.The ability of proliferation in si-NUAK2 group was significantly lower than that in the control group.Transwell and scratch results showed that the migration ability of si-NUAK2 group was significantly lower than that of the negative control group.The results of String database analysis showed that NUAK2 was closely related to proteolysis,physiological rhythm and Wnt signaling pathways.Conclusion NUAK2 may affect the proliferation and migration of human breast cancer cells and slow down the development of breast cancer,which is expected to be a new target for breast cancer treatment.
