摘 要:目的通过生物信息学方法分析Janus激酶3(JAK3)在头颈鳞状细胞癌(HNSCC)中的表达及预后的评估价值。方法从TCGA数据库获取正常组织及HNSCC组织中JAK3的表达数据,分析JAK3在HNSCC中的共表达基因,利用GSEA算法,分析确定JAK3参与的生物学过程及功能采用Survival和Survminer R软件包进行Kaplan-Meier分析,评估预后价值。通过CIBERSORT对免疫细胞浸润进行量化,分析免疫细胞浸润与JAK3表达的关系,JAK3基因与免疫检查点基因表达量的相关性,使用Estimate包对肿瘤样品免疫微环境打分。分别运用TCIA数据库和pRRophetic包评估PD-1、CTLA4免疫治疗的反应和4种化疗药物的疗效。结果JAK3在HNSCC中存在高表达现象,与JAK3的共表达基因主要涉及免疫系统中淋巴细胞CD6、LCK等相关基因,GO富集提示JAK3分布于核糖体,与细胞因子受体相互作用,富集的KEGG信号通路主要与B细胞受体通路、抗原受体介导的信号等通路相关,患者的生存时间与JAK3表达量呈正相关。免疫治疗PD-1对于JAK3高表达组更敏感。结论JAK3在HNSCC中高表达,与HNSCC预后密切相关,并与免疫治疗的疗效相关,可作为HNSCC预后预测的标志物之一,并指导个体化治疗。Objective To analyze the expression and prognostic value of Janus kinase 3(JAK3) in head and neck squamous cell carcinoma(HNSCC) by bioinformatics method.Methods The expression data of JAK3 in normal tissues and HNSCC tissues were obtained from TCGA database.The co-expression genes of JAK3 in HNSCC were analyzed.The biological process and function of JAK3 were analyzed and determined by GSEA algorithm.KaplanMeier analysis was performed by Survival and Survminer R software package to evaluate the prognostic value.CIBERSORT was used to quantify immune cell infiltration,analyze the relationship between immune cell infiltration and JAK3 expression,and the correlation between JAK3 gene and immune checkpoint gene expression.Estimate package was used to score the immune microenvironment of tumor samples.The TCIA database and pRRophetic package were used to evaluate the response to PD-1 and CTLA4 immunotherapy and the efficacy of four chemotherapy drugs,respectively.Results JAK3 was highly expressed in HNSCC,and the co-expression genes of JAK3 were mainly involved in CD6,LCK and other related genes in immune system lymphocytes.GO enrichment suggested that JAK3 was distributed in ribosomes and interacted with cytokine receptors.The enriched KEGG signaling pathway was mainly related to B cell receptor pathway and antigen receptor-mediated signaling pathway,and the survival time of patients was positively correlated with the expression level of JAK3.Immunotherapy PD1 was more sensitive to the group with high JAK3 expression.Conclusions JAK3 is highly expressed in HNSCC,which is closely related to the prognosis of HNSCC and the efficacy of immunotherapy.JAK3 can be used as one of the prognostic markers of HNSCC to guide individual therapy.
