摘  要:目的观察康复训练通过DCC/APPL-1/AKT通路对缺血性脑卒中大鼠功能恢复和神经细胞凋亡的影响并探讨可能作用机制。方法取大鼠100只,将其分为假手术组、自然恢复组、自然恢复+MK2206组、康复训练组、康复训练+MK2206组,每组20只,假手术组不予处理,其余组制作大脑中动脉闭塞模型。建模48 h,自然恢复+MK2206组和康复训练+MK2206组中的大鼠侧脑室注射MK2206试剂,康复训练组、康复训练+MK2206组中的大鼠进行康复训练。于术后2、7、14、21 d进行神经功能和运动功能评分,Western blotting法检测大鼠海马组织DCC、APPL-1、AKT、PAKT蛋白表达,免疫组化染色法检测大鼠海马组织中DCC、APPL-1、AKT阳性表达;流式细胞仪检测大鼠海马神经细胞凋亡和细胞周期。结果与康复训练组相比,自然恢复组、自然恢复+MK2206组和康复训练+MK2206组的神经功能和运动功能评分较高(P均<0.05);与康复训练组相比,自然恢复组、自然恢复+MK2206组和康复训练+MK2206组海马组织中DCC、APPL-1、AKT蛋白表达较低(P均<0.05);与康复训练组相比,自然恢复组、自然恢复+MK2206组和康复训练+MK2206组海马组织中DCC、APPL-1、AKT阳性表达率较低(P均<0.05);与康复训练组相比,自然恢复组、自然恢复+MK2206组和康复训练+MK2206组的凋亡率较高(P均<0.05);与康复训练组相比,自然恢复组、自然恢复+MK2206组和康复训练+MK2206组的G_(0)/G_(1)期较高(P均<0.05)。结论对于缺血性脑卒中大鼠,康复训练可通过DCC/APPL-1/AKT信号通路,抑制神经细胞凋亡,有助于缺血性脑卒中大鼠神经和运动功能恢复。Objective To observe the effects of rehabilitation training on functional recovery and neuronal apoptosis in rats with ischemic stroke through DCC/APPL-1/AKT pathway and to investigate its mechanism.Methods Totally 100 rats were divided into the sham operation group,natural recovery group,natural recovery+MK2206 group,rehabilitation training group,and rehabilitation training+MK2206 group.Except the sham operation group,rats in the other groups were used to make ischemic stroke models.At 48 h of modeling,rats in the natural recovery+MK2206 group and rehabilitation training+MK2206 group were given MK2206 reagents through intracerebroventricular injection,and rats in the rehabilitation training group,rehabilitation training+MK2206 group received rehabilitation training.The neural function and motor function were scored on the 2nd,7th,14th,and 21st days after operation.The expression of DCC,APPL-1,AKT,PAKT protein in rat hippocampus was detected by Western blotting,and the positive expression of DCC,APPL-1,and AKT protein in rat hippocampus was detected by immunohistochemistry.The apoptosis and cell cycle of hippocampal neurons were detected by flow cytometry.Results Compared with the rehabilitation training group,the scores of nerve function and motor function in the natural recovery group,natural recovery+MK2206 group and rehabilitation training+MK2206group were higher(all P<0.05).Compared with the rehabilitation training group,the DCC,APPL-1 and AKT proteins in the hippocampus of the natural recovery group,the natural recovery+MK2206 group and the rehabilitation training+MK2206 group were lower(all P<0.05).Compared with the rehabilitation training group,the positive expression rates of DCC,APPL-1 and AKT in hippocampus of natural recovery group,natural recovery+MK2206 group and rehabilitation training+MK2206 group were lower(all P<0.05).Compared with the rehabilitation training group,the apoptosis rates of natural recovery group,natural recovery+MK2206 group and rehabilitation training+MK2206 group was higher(all
