摘  要:为了探讨不同来源和剂量的β-葡聚糖(BG)对小鼠体重和主要器官组织病理学的影响,试验将70只小鼠随机分为对照组、20 mg/kg酵母BG组、50 mg/kg酵母BG组、20 mg/kg细菌BG组、50 mg/kg细菌BG组、20 mg/kg藻类BG组和50 mg/kg藻类BG组,每组10只。对照组灌胃生理盐水,其余各BG组每天分别灌胃相应来源和剂量的BG,连续14 d。期间记录小鼠的临床症状,于第14天处死小鼠,采集心脏、肝脏、脾脏、肺脏、肾脏、十二指肠、空肠和回肠,进行H.E.染色并观察组织病理学变化。结果表明:试验第14天,除50 mg/kg细菌BG组外,其余各BG组小鼠体重均极显著低于对照组(P<0.01)。同时BG来源和剂量对小鼠体重有极显著交互作用(P<0.01)。光镜下对照组小鼠各器官组织未见明显病理变化。各BG组部分小鼠可见心脏组织心肌纤维变性,肝脏组织中央静脉周围肝细胞变性,脾脏组织红髓内中性粒细胞增多,肺脏组织肺泡上皮细胞坏死、炎性细胞浸润,十二指肠、空肠和回肠组织肠绒毛坏死等病理变化,肾脏组织未见明显病理损伤。说明BG在小鼠中的安全使用剂量为20 mg/kg以内,超过20 mg/kg会明显降低小鼠体重,引起组织器官尤其肺脏组织的病理损伤。In order to explore the effects of different β-glucan(BG) sources and doses on body weight and histopathology of major organs in mice, seventy mice were randomly divided into control group, 20 mg/kg yeast BG group, 50 mg/kg yeast BG group, 20 mg/kg bacterial BG group, 50 mg/kg bacterial BG group, 20 mg/kg algae BG group and 50 mg/kg algae BG group, with 10 mice in each group. The control group was given normal saline by gavage, and the other groups were given the corresponding dose and source of BG by gavage every day for 14 consecutive days. During the period, the clinical symptoms of the mice were recorded. The mice were sacrificed on the 14th day. The heart, liver, spleen, lung, kidney, duodenum, jejunum and ileum were collected for H.E. staining and histopathological changes were observed. The results showed that except for 50 mg/kg bacterial BG group on the 14th day of the experiment, the body weight of mice in the other BG groups was significantly lower than that in the control group(P<0.01). There was a significant interaction in the effects of the source and dose of BG on body weight in mice(P<0.01). There was no obvious pathological changes in the organs and tissues of the animals in the control group under the light microscope. Some animals in the BG group showed myocardial fiber degeneration in the heart tissue, degeneration of hepatocytes around central vein in the liver tissue, the increased number of of red pulp neutrophils in the spleen tissue, necrosis of alveolar epithelial cells and inflammatory cell infiltration in the lung tissue, necrosis of intestinal villi in the duodenum, jejunum and ileum, but no obvious pathological changes was found in the kidney tissue. The results indicated that the safe dosage of BG in mice should be less than 20 mg/kg, and more than 20 mg/kg would significantly reduce the body weight of mice and cause pathological changes of tissues and organs, especially lung tissue.
