摘  要:目的:基于16S rDNA技术,探讨生姜汁(ZOS)对脂多糖诱导的急性肺损伤(ALI)小鼠的影响。方法:将Balb/c小鼠分为对照组、模型组和ZOS组,其中对照组和模型组灌胃生理盐水,ZOS组灌胃ZOS(3 g/mL),连续给药14 d后,LPS滴鼻制备急性肺损伤小鼠模型,12 h后取小鼠粪便应用16S rDNA测序分析技术检测。结果:在门水平上,小鼠肠道内微生物优势菌群为厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidota),在目水平上排名前4位的为拟杆菌目(Bacteroidales)、乳螺目(Lachnospirales)、梭菌目UCG-014(Clostridia_UCG-014)、振螺目(Oscillospirales)。样本中细菌群落组成中包括19种菌属,其中ZOS组Muribaculaceae、Lachnospiraceae、普雷沃氏菌科UCG-001、拟杆菌属、拟普雷沃氏菌属、Oscillospiraceae、Clostridia_vadinBB60_group、Ruminococcaceae菌群丰度有上升趋势,Clostridia_UCG-014、毛螺菌科NK4A136、罗氏菌属、Lachnospiraceae、嗜木聚糖真杆菌的菌群丰度有明显下降趋势。结论:给予ZOS后可改善小鼠肠道微生物多样性,对LPS诱导的急性肺损伤具有明显治疗作用。【Objective】 The effect of ZOS on acute lung injury(ALI) induced by Lipopolysaccharide(LPS) in mice was investigated by 16S rDNA technique.【Method】 mice were divided into control group, model group and ZOS group.ZOS group was given 3 g/mL ZOS by gavage, and control group and model group were given the same volume of normal saline once a day for consecutive 14 days.Mice model of acute lung injury was prepared by LPS intranasal drip.Feces were collected 12 h later and analyzed by 16S rDNA sequencing. 【Result】 At phylum level, the dominant microflora in the intestinal tract of mice were Firmicutes and Bacteroidota.In order level, the top four were Bacteroidales, Lachnospirales, Clostridia_UCG-014 and Oscillospirales.The bacterial community composition in the samples included 19 genera.The intestinal flora with increasing abundance in ZOS group included Muribaculaceae, Lachnospiraceae and Prevoelsiaceae UCG-001,Bacteroides,Prevotella,Oscillospiraceae,Clostridia_vadinBB60_group,Ruminococcaceae.The abundance of clostridia_UCG-014,NK4A136 of Spirillaceae,Roxella,Lachnospiraceae and Eubacter xylanophilus decreased significantly.【Conclusion】 ZOS can regulate the intestinal microbial diversity of mice and has obvious therapeutic effect on LPS-induced acute lung injury.
