摘  要:通过单因素试验优化了耐盐人苍白杆菌(Ochrobactrum anthropi)N2-2的脱氮发酵培养条件。利用Plackett-Burman试验得出影响菌株N2-2氨氮降解效率最显著的3个因素为pH、丁二酸钠质量浓度和温度。通过Central Composite Design试验设计对菌株N2-2的培养条件进行响应面优化并分析得出最佳脱氮发酵培养条件。结果表明,在丁二酸钠质量浓度为9.1 g/L、初始氨氮质量浓度为300 mg/L、pH为8.6、转速为120 r/min、温度为30℃、接种量为4%的条件下,氨氮降解率为84.97%,比优化前提高了11.78%。The denitrification and fermentation conditions for salt-tolerant Ochrobactrum anthropi N2-2 are optimized by single factor experiment.Plackett-burman experiment is utilized to determine three most significant factors affecting the degradation efficiency of ammoniacal nitrogen by strain N2-2,including pH,sodium succinate and temperature.Response surface optimization is carried out for the culture conditions of strain N2-2 through Central Composite Design, and the optimal denitrification fermentation culture conditions are obtained as follows: the concentration of sodium succinate is 9.1 g·L-1,the initial mass concentration of ammoniacal nitrogen is 300 mg·L-1,pH=8.6,the rotation speed is 120 r·min-1,the temperature is 30℃,and the inoculation amount is 4%.Under these conditions, the degradation rate of ammoniacal nitrogen reaches 84.97%,11.78% higher than before optimization.
