摘　　要：目的探讨同源异形盒基因A1(HOXA1)在结肠癌中的表达及临床意义。方法随机选取2021年3~10月宁夏医科大学总医院结直肠外科收治的结肠癌患者作为研究对象,运用RT-qPCR检测66例冰冻新鲜结肠癌组织及配对正常组织中HOXA1基因mRNA的表达;运用免疫组织化学检测203例结肠癌石蜡组织芯片中HOXA1蛋白的表达水平,并分析其与临床病理参数间的关系。结果RT-qPCR结果显示,结肠癌中HOXA1 mRNA的相对表达量为5.634±0.563,明显高于正常结肠组织的1.893±0.442,差异具有显著统计学意义(P<0.01);免疫组织化学结果显示,HOXA1蛋白主要表达于细胞核中,其在结肠癌中的表达阳性率为62.56%,明显高于正常组织的21.67%,差异具有显著统计学意义(P<0.01);结肠癌中HOXA1表达水平与AJCC分期、T分期、淋巴结转移、远处转移及组织学分型均有关(P<0.05)。结论HOXA1在结肠癌中异常高表达,与肿瘤分期淋巴结转移等高危因素相关,是结肠癌潜在治疗靶点。Objective To investigate the expression pattern of homeobox A1(HOXA1)and its clinical signifi-cance in colon cancer.Methods Colon cancer patients admitted to Department of Anal-colorectal Surgery,the General Hospital of Ningxia Medical University from March 2021 to October 2021 were selected as the research objects.RT-qP-CR was used to detect the expression of HOXA1 gene mRNA in 66 fresh-frozen paired specimens of colon cancer tis-sues and normal tissues.Immunohistochemistry was used to detect the HOXA1 protein expression in 203 cases of colon cancer by paraffin tissue microarray,and the correlation between HOXA1 expression and clinicopathological features was analyzed.Results RT-qPCR results showed that the average mRNA expression of HOXA1 in colon cancer was 5.634±0.563,significantly higher than 1.893±0.442 in normal tissue,respectively.Results of immunohistochemistry indi-cated that HOXA1 mainly expressed in nucleus of colon cancer cells,and the expression level was higher in colon can-cer tissue than that in the corresponding normal colon tissue(62.56%vs 21.67%,P<0.01).Overexpression of HOXA1 was significantly correlated with American Joint Committee on cancer(AJCC)stage,T stage,lymph node metastasis,distant metastasis,and histological classification(P<0.05).Conclusion HOXA1 is significantly overexpressed in colon cancer patients.The abnormal expression of HOXA1 is correlated with a series of high risk factors of colon cancer such as tumor staging and lymph node metastasis,which might be a potential target for colon cancer therapy.
