一株致种鸭瘸腿的肠炎沙门氏菌分离与鉴定

摘　　要：为探究育成期种鸭瘸腿的发生原因,采集病鸭的肝脏、关节进行病原分离,经沙门氏菌stn基因PCR、多重PCR及7对管家基因MLST分析等方法进行鉴定,并对分离菌进行生化试验、药敏试验和SPF鸭致病性试验。结果显示:4份肝脏及关节组织病毒检测为阴性、细菌检测为肠炎沙门氏菌阳性,PCR、MLST及生化试验结果确定分离了一株肠炎沙门氏菌;该分离菌对新霉素和多西环素药物较为敏感;SPF鸭致病性试验结果显示该分离菌具有较强的致病性,可复制出与临床表现一致的纤维素性心包炎、肝周炎症状,感染鸭瘸腿比例为100%,剖检有严重的腿肌干酪样物。研究首次报道了肠炎沙门氏菌感染可引起种鸭瘸腿和严重的纤维素性心包炎、肝周炎、腿肌干酪样物,为育成期种鸭瘸腿的防治提供参考。In order to explore the causes of lameness in breeding ducks, the pathogen was isolated and cultured from liver and joints of diseased breeder duck, identified by PCR for stn gene of Salmonella, multiplex PCR and MLST analysis for seven pairs of housekeeper genes, and the biochemical test, drug sensitivity test and SPF duck pathogenicity test were conducted. The results showed that a strain of Salmonella Enteritidis was isolated from liver and joints samples with negative detection of viruses.The PCR, MLST and biochemical test results were confirmed that the isolate was a strain of Salmonella Enteritidis. The isolate was more sensitive to neomycin and doxycycline. SPF duck pathogenicity test showed that the isolate had strong pathogenicity,and could reproduce fibrinous pericarditis and perihepatic inflammation which were consistent with the clinical feature, the proportion of infected ducks with lameness was 100%, and there were serious caseous substance in leg muscle in autopsy. This study reported for the first time that the infection of Salmonella Enteritidis could cause lameness and severe fibrinous pericarditis, perihepatic inflammation and caseous substance in leg muscle of breeding duck, which provided a reference for treatment of lameness of breeder ducks in finishing period.
