摘  要:试验旨在研究脱氧雪腐镰刀菌烯醇(DON)对蛋鸡肠道形态结构、炎症因子、紧密连接蛋白表达的影响。选取26周龄产蛋率一致的海兰褐蛋鸡80只,随机分为4组,每组4个重复,每个重复5只,对照组每天灌服等量生理盐水,试验组每天分别灌服1、5和10mg/kg·BW的DON,连续灌服6周。结果显示:与对照组相比,5和10mg/kgDON组肠道形态结构均受到不同程度损伤,出现黏膜脱落、绒毛数量减少的现象,绒毛长度显著降低,隐窝深度增加,且呈剂量-效应关系(P<0.05或P<0.01)。DON剂量≥5mg/kg时,白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、紧密连接蛋白-1(ZO-1)和紧密连接跨膜蛋白(claudin-1)的基因表达水平降低(P<0.05或P<0.01);DON剂量为10mg/kg时,紧密连接蛋白-1(ZO-1)和紧密连接跨膜蛋白(claudin-1)的蛋白表达也显著降低(P<0.05);DON剂量为1mg/kg时,白细胞介素8(IL-8)表达水平显著增加(P<0.01)。表明DON高于5mg/kg·BW时能显著破坏蛋鸡肠道形态、降低紧密连接蛋白、抑制肠道免疫,导致蛋鸡肠道屏障完整性破坏和功能损伤。The aim of this study was to investigate the effects of deoxynivalenol(DON) on intestinal morphological structure,inflammatory factors and tight junction protein expression in laying hens. Eighty 26-week-age Hy-Line Brown laying hens with consistent egg production rate were randomly divided into 4 groups with 4 replicates and 5 hens per replicate. The laying hens were respectively given 0, 1, 5 and 10 mg/kg·BW DON daily by intragastric administration for 6 weeks, respectively. The results showed that the damage of microvilli exfoliated and reduction of villous number in duodenum were observed after exposure to DON. Compared with control group, the villous length was significantly decreased and the crypt depth was remarkably increased in 5 and10 mg/kg DON group in a dose-effect relationship(P<0.05 or P<0.01), the expression levels of interleukin 6(IL-6), interleukin β(IL-1β), and claudin-1 were significantly reduced(P<0.05 or P<0.01). The expression level of tight junction protein 1(ZO-1)and claudin-1 was significantly reduced when the dose of DON was 10 mg/kg(P<0.05). Furthermore, the expression level of interleukin 8(IL-8) was significantly increased in 1 mg/kg DON group than that in control group(P<0.01). It was concluded that when DON was more than 5 mg/kg, it could significantly disrupt the intestinal morphology, reduce tight junction proteins, inhibit intestinal immunity, and ultimately lead to the destruction of intestinal barrier integrity and functional impairment in laying hens.
