摘  要:该试验通过研究洋甘菊残渣的成分及抑菌活性来探究洋甘菊残渣的应用价值。通过对各极性部位中的酚酸、多糖、黄酮类化合物的鉴定及含量测定,初步判定有效抑菌物质;采用打孔法和试管二倍稀释法测定各极性部位对金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌、绿脓杆菌的抑菌活性;探究洋甘菊残渣中乙酸乙酯萃取物对金黄色葡萄球菌的生长曲线、电导率、细胞壁完整性、胞外多糖、胞外蛋白的影响。结果表明,洋甘菊残渣各极性部位中均含有黄酮类化合物,其中乙酸乙酯萃取物中黄酮类化合物含量最高,为31.53 mg/g;乙酸乙酯萃取物对金黄色葡萄球菌的抑制作用最强,最小抑菌浓度(minimal inhibitory concentration,MIC)为15.63 mg/mL,抑菌圈直径为(19.73±0.45)mm;抑菌机制试验表明,金黄色葡萄球菌经乙酸乙酯萃取物处理后,细胞中的内容物流出,胞外蛋白、多糖含量显著升高,菌体培养液电导率也显著增大。This experiment was conducted to investigate the application value of chamomile residues by analyzing the composition and antibacterial activity of chamomile residues.Through the identification and content determination of phenolic acid,polysaccharides,and flavonoids in each polar site,this paper preliminarily determined the effective antibacterial substances.The antibacterial activity of each polar part against Staphylococcus aureus,Escherichia coli,Bacillus subtilis,and Pseudomonas aeruginosa was determined by using the perforation method and tube double dilution method.The effects of ethyl acetate extract from chamomile residue on the growth curve,electrical conductivity,cell wall integrity,extracellular polysaccharides,and extracellular protein of S.aureus were explored.The results showed that all the polar parts of chamomile residue contained flavonoids,among which the ethyl acetate extract contained the highest amount of flavonoids(31.53mg/g).The ethyl acetate extract had the strongest antibacterial effect on S.aureus with the minimum inhibitory concentration(MIC)of 15.63 mg/mL.The diameter of the inhibition circle was(19.73±0.45)mm.The antibacterial mechanism test showed that after the treatment of S.aureus with ethyl acetate extract,the content of the cells flowed out,the extracellular protein and polysaccharide content increased significantly,and the conductivity of the culture solution of the bacteria also increased significantly.
