摘 要:目的探讨七叶皂苷钠(sodium aescinate,SA)对多柔比星(doxorubicin,DOX)抑制人乳腺癌细胞MCF-7生长和转移作用的影响及机制。方法将人乳腺癌细胞MCF-7设置对照组及不同药物处理组,通过MTT检测MCF-7细胞增殖活性并选定最合适的处理时间及药物浓度。细胞克隆形成实验及Transwell实验检测不同药物处理对细胞增殖、迁移及侵袭能力的影响;实时荧光定量PCR(quantitative real-time-PCR,qRT-PCR)及免疫蛋白印迹检测上皮-间质转化指标上皮型钙黏蛋白、神经型钙黏蛋白和波形蛋白在mRNA和蛋白水平的表达及PI3K/Akt通路蛋白表达水平。结果MTT结果示联合使用DOX以较低的浓度0.25μM可达到半数抑制率。克隆形成实验结果同样提示联合处理时细胞克隆数仅占对照组的50%。Transwell实验结果表明联合药物处理MCF-7细胞,细胞迁移和侵袭细胞数较对照组下降70%。上皮型钙黏蛋白在mRNA和蛋白水平增加,反之减少神经型钙黏蛋白、波形蛋白的表达。结论SA能够协同DOX通过PI3K/Akt信号通路抑制人乳腺癌细胞MCF-7的生长和转移。Objective To explore the effect and mechanism of sodium aescinate(SA)on doxorubicin(DOX)inhibiting the growth and metastasis of MCF-7 breast cancer cell.Methods MCF-7 cell were divided into control group and different drug treatment groups.The proliferation activity of MCF-7 cell was measured by MTT assay.The effects of different drug treatments on cell proliferation,migration and invasion were detected by cell clone formation assay and Transwell assay.Real-time quantitative PCR(qRT-PCR)and Western blot were used to detect the expression of E-cadherin,N-cadherin and Vimentin at mRNA and protein levels.The expression levels of PI3K/Akt pathway were detected by western blot.Results DOX at a lower concentration of 0.25μM could achieve half inhibition rate in MTT assay.The results of colony formation experiment also suggested that the number of cell clones was only 50%in the control group.Transwell experiments showed that the number of migration and invasion cells of MCF-7 cells decreased by 70%compared with the control group.E-cadherin increased the mRNA and protein levels and decreased the expression of N-cadherin and Vimentin.Conclusion SA can improve the effect of DOX of MCF-7 breast cancer cell through PI3K/Akt signaling pathways,further inhibit the proliferation and metastasis of MCF-7 breast cancer cell.
