摘 要:目的:研究绿原酸对不同水平环二鸟苷酸(c-di-GMP)铜绿假单胞菌(P.a)早期和成熟生物被膜(BF)的抑制作用。方法:用液体培养基稀释法测定最低抑菌浓度(MIC)。构建3种P.a菌株—PAO1、P.a胞内c-di-GMP高表达菌株(PAO1ΔwspF)和P.a胞内c-di-GMP低表达菌株(PAO1/Plac-yhjH)BF体外模型,分别分为空白对照组和不同浓度的绿原酸(1/2 MIC、1/4 MIC、1/8 MIC)组。结晶紫染色半定量法测定3 d及7 d载体表面BF总量,连续稀释法计数绿原酸作用3 d及7 d后BF内活菌,观察绿原酸对运动能力的影响。结果:在绿原酸的作用下,无论3 d还是7 d载体表面所形成BF均少于空白对照组(P<0.05),BF内活菌均较空白对照组少(P<0.05),3种菌株的运动区域直径均小于空白对照组(P<0.05)。结论:绿原酸对不同水平c-diGMP P.a早期和成熟BF均具有抑制作用,对BF内活菌具有良好的杀灭效果,影响其运动能力。Objective:To study the inhibitory effects of chlorogenic acid on the early and mature biofilm (BF) of Pseudomonas aeruginosa (P.a) at different levels of cyclic diguanosine (c-di-GMP).Methods:The minimum inhibitory concentration (MIC) was determined by liquid medium dilution method.BF models of three P.a strainsPAO1,P.a strain with high expression of intracellular c-di-GMP (PaO1ΔwspF) and P.a strain with low expression of intracellular c-di-GMP (PAO1/Plac-yhjH) were constructed in vitro and the models were divided into blank control group and chlorogenic acid (1/2 MIC,1/4 MIC,1/8 MIC) group with different concentrations.The tota amount of BF on the carrier surface of 3 d and 7 d was determined by semi-quantitative crystal violet staining and the viable bacteria in BF after 3 and 7 days of chlorogenic acid treatment were counted by continuous dilution method to observe the effects of chlorogenic acid on exercise ability.Results:Under the action of chlorogenic acid,the BF formed on the carrier surface of both 3d and 7d was less than that of the blank control group (P<0.05),the number of viable bacteria in BF was less than that of the blank control group (P<0.05),and the diameter of the moving area of the three strains was smaller than that of the blank control group (P<0.05).Conclusion:Chlorogenic acid can inhibit early and mature BF of P.a at different levels of c-di-GMP and has a good killing effect on BF live bacteria,affecting their exercise ability.
