摘  要:目的研究菊苣总黄酮(CTF)对酒精所致小鼠急性肝损伤的保护作用.方法小鼠被随机分为空白对照组(纯净水)、酒精模型组(纯净水)、阳性对照组(西利马林,10 mg/kg bw)、CTF低剂量组(50 mg/kg bw)和高剂量组(100 mg/kg bw).除空白对照组外,其余各剂量组在最后一次灌胃处理后禁食6 h,继而均灌胃56度红星二锅头白酒(12 mL/kg bw).实验结束后,采集所有小鼠血浆和肝脏,测定血浆相关指标,肝脏分别用于组织病理分析、生化分析及实时荧光定量PCR分析.结果CTF高剂量组能够明显缓解由酒精引起的肝脏指数即血浆ALT和AST活性的上升.CTF低剂量组和高剂量组血浆TC和TG水平显著降低.此外,CTF高剂量组肝脏中MDA水平明显降低,而GSH、GSH-PX和SOD水平有不同程度的升高.CTF高剂量组肝脏Nrf-2和HO-1基因的mRNA表达水平显著升高,而CYP2e1、TNF-α、TLR-4和FAS基因的表达水平则明显降低.结论菊苣总黄酮可以提高肝脏的抗氧化和抗炎能力,对酒精所致的急性肝损伤有明显的缓解作用.Objective To explore the effect of Chicory total flavonoids(CTF)on alcohol-induced acute liver injury in mice.Methods Mice were randomly divided into blank control group(purified water),alcohol-induced liver injurymodel group(purified water),positive control group(silymarin 10 mg/kg bw),CTF low-dose group(50 mg/kg bw)and the high-dose group(100 mg/kg bw).Except the blank control group,each group of mice was fasted for 6 h after the last treatment and was intragastrically administrated with 56%Red Star Erguotou liquor(12 mL/kg bw).At the end of experiment,all mouse serum and liver were collected,serum-related indicators and liver-related indicators were determined,A portion of the liver was used for histopathological analysis,whereas the rest was used for biochemical analysis and real-time PCR analysis.Results The high-dose groups of CTF significantly alleviated the alcohol-induced increases in liver index,serum ALT and AST activities.Serum TC and TG level were significantly reducedin both the low-dose and high-dose CTF groups.Furthermore,MDA contentof the high-dose CTF groupin liver was significantly reduced,accompanying with the levels of GSH,GSH-PXand SOD elevated to various extents.The mRNA expression levels of hepatic Nrf-2 and HO-1 genes were significantly increased in the high-dose group of CTF,while those of CYP2e1,TNF-α,TLR-4 and FAS were significantly decreased.Conclusion Chicory total flavonoids could improve the antioxidant and anti-inflammatory capacities of the liver,and has significantly relieving effect on alcohol-induced acute liver injury inmice.
