摘  要:为了解析碱液预处理对柠条锦鸡儿茎理化结构及酶解的影响,实现柠条锦鸡儿的高值化利用,文章比较了柠条锦鸡儿叶和不同径级茎的可消化性,并分析了NH_(3)·H_(2)O,Ca(OH)_(2)和NaOH预处理对柠条锦鸡儿茎的化学组分、化学基团、结晶度、孔径分布和酶解还原糖得率的影响。研究结果表明:柠条锦鸡儿叶和细茎(直径<0.5 cm)是优良的饲用原料;碱液预处理能够降解柠条锦鸡儿茎中的半纤维素和中性洗涤物,能够断裂木质素与半纤维素间的酯键以及木质素间的醚键;随着化学结构的改变,柠条锦鸡儿茎的致密结构被破坏,孔隙率增大,纤维素结晶区暴露;与对照组相比,NH_(3)·H_(2)O,Ca(OH)_(2)和NaOH预处理后的酶解还原糖得率分别提高了66.68%,17.91%,93.12%。碱液预处理能有效改变柠条锦鸡儿茎的理化结构,提高酶解还原糖得率,是柠条锦鸡儿等荒漠灌木的理想预处理方式。To realize high-value utilization of Caragana korshinskii and understand the effects of alkali pretreatment on the physical and chemical structure and enzymatic hydrolysis of stem from Caragana korshinskii,the digestibility of stem and leaf from Caragana korshinskii was compared,additionally,the medium and thick stem(diameter greater than 0.5 cm)of Caragana korshinskii further pretreated by NH_(3)·H_(2)O,Ca(OH)_(2)and NaOH,in sequence,and the effects of alkali pretreatment on the chemical composition,chemical groups,crystallinity,pore size distribution,accessibility and reducing sugar yield were investigated.The results showed that the leaf and thin stem(diameter lower than 0.5 cm)of Caragana korshinskii exhibited excellent forage quality.Alkali pretreatment could degrade hemicelluloses and neutral detergent soluble,and break down the ester bonds between cellulose and lignin,and the ether bonds within lignin,yet,without obvious degradation of lignin.With the destruction of rigid structure and exposure of cellulose,the reducing sugar yield of samples pretreated by NH_(3)·H_(2)O,Ca(OH)_(2)and NaOH was increased by66.68%,17.91%and 93.12%,respectively.Alkali pretreatment is an ideal method for desert shrubs such as Caragana korshinskii,which can destroy the physicochemical structure to improve enzymatic digestibility under mild conditions.
