摘 要:咖啡酰辅酶A氧甲基转移酶(caffroyl-CoA O-methyltransferase,CCoAOMT)是一类S-腺苷-L-甲硫氨酸(SAM)甲基转移酶,本研究采用生物信息学方法,旨在鉴定和注释刺梨中CCoAOMT家族基因的全基因组序列,分析RrCCoAOMT基因的分布,同时分析RrCCoAOMT基因结构与其启动子顺式调控元件。结果表明,刺梨中预测具有3个CCoAOMT基因,氨基酸的长度范围为180~245,分子量范围为20496.53~27815.17 kD,等电点为4.70~6.14。蛋白质二级结构预测结果表明RrCCoAOMT家族蛋白主要以α-螺旋和不规则卷曲为主。本研究对CDS上游2 kb启动子区域的顺式元件分析表明,光是影响该家族功能的重要因素。亚细胞定位预测表明RrCCoAOMT家族蛋白主要定位于细胞质中,其中仅CCoAOMT01定位到细胞骨架上,预测其与木质素的合成最相关。RT-qPCR结果表明:经遮阴处理后,CCoAOMT01的基因表达量逐渐下降,而外源Ca^(2+)处理则上调CCoAOMT01的表达;经ETH及SA处理后,CCoAOMT01的基因表达量表达模式为先下降,后上升。以上结果为进一步研究刺梨CCoAOMT参与木质素合成机制提供了理论参考。Caffeoyl-CoA O-methyltransferase(CCoAOMT)is a kind of S-adenosine-L-methionine(SAM)methyltransferase.In this study,bioinformatics methods were used to identify and annotate the whole genome sequence of CCoAOMT family genes in Rosa roxburghii,analyze the distribution of RrCCoAOMT genes,and analyze the gene structure of RrCCoAOMT and its promoter cis-regulatory elements.The results showed that there were three CCoAOMT genes predicted in Rosa roxburghii,the length of amino acids ranged from 180 to 245,the molecular weight ranged from 20496.53~27815.17 kD,and the isoelectric point ranged from 4.70 to 6.14.The prediction results of protein secondary structure showed that the protein of RrCCoAOMT family was mainly composed ofαhelix and random coil.Cis element analysis of 2 kb promoter region upstream of CDS showed that light was an important factor affecting the function of this family.Subcellular localization prediction showed that RrCCoAOMT family proteins were mainly located in cytoplasm,and only CCoAOMT01 was located on cytoskeleton,which predicted that it was most related to lignin synthesis.The results of RT-qPCR showed that after shading treatment,the gene expression of CCoAOMT01 decreased gradually,while exogenous Ca^(2+)treatment increased the expression of CCoAOMT01.After ETH and SA treatment,the gene expression pattern of CCoAOMT01 decreased first and then increased.The above results provide a theoretical reference for further study on the mechanism of Rosa roxburghii participating in lignin synthesis.
