摘  要:为了解辣椒愈伤组织形成及影响辣椒素积累的条件,本研究以‘杭椒五号’和‘文昌小米椒’为实验材料,探究了愈伤组织形成的影响因素,并对不同光照和氯化钠浓度处理下愈伤组织中辣椒素类化合物的含量进行了测定和比较。结果表明:以下胚轴为外植体,在MS+2.0 mg/L 2,4-D+0.5 mg/L 6-BA激素配比下,愈伤组织诱导效果最佳;相较于黑暗条件,16 h光照8 h黑暗条件下,‘文昌小米椒’和‘杭椒五号’辣椒素含量分别提升183.5%和1268.7%,生长指数分别提升36.0%和38.5%;3.0 g/L氯化钠处理下,‘文昌小米椒’和‘杭椒五号’辣椒素积累达到最大,分别提升299.9%和477.2%,氯化钠对‘文昌小米椒’生长指数无显著影响,但1.0 g/L氯化钠可以使‘杭椒五号’愈伤组织生长指数提升2.54倍;光照和氯化钠同时处理,辣椒素含量较单独光照处理要低,但促进了愈伤组织的生长。氯化钠和光照均是促进辣椒愈伤组织产生辣椒素的关键影响因素,本研究将对相关研究人员探究辣椒素代谢调控和体外辣椒素高效生产提供借鉴。In order to understand the conditions of pepper callus formation and capsaicin content,this study used’Hangjiao No.5’and’Wenchang millet pepper’as experimental materials to explore the influencing factors of callus formation and the influence of different light and sodium chloride concentration on the accumulation of capsaicinoid compounds in callus.The results showed that the hypocotyls were explants,and the callus induction effect was the best under the ratio of MS+2.0 mg/L 2,4-D+0.5 mg/L 6-BA hormone.Compared with dark conditions,under 16 h light and 8 h dark conditions,the accumulation of capsaicin in’Wenchang millet pepper’and’Hangjiao No.5’increased by 183.5%and 1268.7%,and the growth index increased by 36.0%and 38.5%.Under the treatment of 3.0 g/L sodium chloride,the accumulation of capsaicin in’Wenchang millet pepper’and Hangjiao No.5reached the maximum,increasing by 299.9%and 477.2%respectively.Sodium chloride has no significant effect on the growth index of’Wenchang millet pepper’,but 1.0 g/L sodium chloride can increase the callus growth index of’Hangjiao No.5’by 2.54 times.Treatment of light and sodium chloride at the same time,the content of capsaicin was lower than that of light treatment alone,but it promoted the growth of callus.Both sodium chloride and light are the key factors that promote the production of capsaicin from pepper callus.This study will provide reference for relevant researchers to explore the regulation of capsaicin metabolism and efficient production of capsaicin in vitro.
