摘 要:目的:建立乳铁蛋白介导的阿霉素纳米脂质载体的制备方法,并对其进行表征。方法:通过对制备条件、表面活性剂种类、脂质材料及有机溶剂等单因素考察,采用乳化-溶剂挥发法制备阿霉素纳米脂质载体;用碳化二亚胺偶联法,将乳铁蛋白连接到纳米脂质载体表面,制备载阿霉素-乳铁蛋白配体纳米脂质载体;对阿霉素纳米脂质载体及乳铁蛋白介导的阿霉素纳米脂质载体形态学特性通过透射电子显微镜(TEM)检测,并采用激光散射粒径测定仪测定其粒径分布、Zeta电位和PDI。结果:根据优化工艺制备出阿霉素纳米脂质载体,并成功将乳铁蛋白偶联至该纳米脂质载体表面;外观形态良好,分散性较好,粒度较为均匀;空白纳米脂质载体粒径在194 nm左右,载药后,粒径有所增大,但变化不明显,当载药的纳米脂质载体连接上乳铁蛋白后,粒径增大到241.0 nm。结论:从TEM形态学观察和粒径的变化上均能证明乳铁蛋白的成功连接,本研究成功制备了乳铁蛋白介导的阿霉素纳米脂质载体,有望成为载阿霉素的新型肺癌靶向制剂。Objective:To establish a method for the preparation and characterization of lactoferrin-mediated doxorubicin nanostructured lipid carriers.Methods:Doxorubicin nanostructured lipid carriers were prepared by emulsification solvent evaporation method with investigating the preparation conditions,surfactant types,lipid materials and organic solvents.Lactoferrin was attached to the surface of nanostructured lipid carrier by carbodiimide coupling method to prepare adriamycin lactoferrin ligand loaded nanostructured lipid carrier.The morphological characteristics of doxorubicin nanostructured lipid carrier and lactoferrin mediated doxorubicin nanostructured lipid carrier were detected by transmission electron microscope(TEM),and the particle size distribution,zeta potential and PDI were measured by laser scattering particle size analyzer.Results:Doxorubicin nanostructured lipid carrier was prepared according to the optimized process,and lactoferrin was successfully coupled to the surface of thenanostructured lipid carrier.Their appearance and dispersion were good,and particle size was uniform.The particle size of blank nanostructured lipid carrier was about 194nm.After drug loading,the particle size increased,but the change was not obvious.When the drug loaded nanostructured lipid carrier was connected with lactoferrin,the particle size increased to 241.0 nm.Conclusion:The morphological observation of TEM and the change of particle size can prove the successful connection of lactoferrin.In this study,lactoferrin-mediated doxorubicin nanostructured lipid carrier was successfully prepared,which is expected to be a new type of lung cancer targeting preparation of containing doxorubicin.
