摘 要:目的:探究胚胎期双酚A(BPA)暴露对斑马鱼眼结构及视觉功能的影响,为环境污染物暴露致子代视觉损害提供理论依据。方法:构建胚胎期BPA暴露斑马鱼模型,体视显微镜观察并测量幼鱼的眼面积、眼长、体长;HE染色检测幼鱼视网膜结构变化;斑马鱼行为学检测系统评估幼鱼视觉行为。结果:(1)50μmol·L-1BPA暴露后幼鱼眼面积、眼身长比显著减小。(2)BPA暴露后幼鱼视网膜细胞排列拥挤,分层紊乱,感光细胞层细胞排列不齐。(3)受精后24 h(24 hpf)BPA暴露组的斑马鱼胚胎摆尾活动减少;96、120 hpf幼鱼动眼频率和幅度降低;120、144 hpf幼鱼自主运动能力显著下降,游泳距离显著缩短。结论:胚胎期BPA暴露可造成斑马鱼眼结构的改变和异常的视觉行为,说明环境污染物会影响子代视觉的发育,提示儿童眼保健应从孕期保健开始。Objective:To explore the effects of exposure to bisphenol A(BPA)on eye structure and visual function in zebrafish embryos,and to provide theoretical basis for the visual damage of offspring caused by exposure to environmental pollutants.Methods:Zebrafish models exposed to different concentrations of BPA in embryonic stage were constructed.Stereomicroscope was used to observe and measure the eye area,eye length and body length of deformities of larvae,zebrafish behavioral detection system was employed to detect the visual behavior of larvae,and HE staining was used to detect the structural changes of retina.Results:(1)After exposure to 50μmol·L-1BPA,the ratio of eye area and eye length zebrafish offspring decreased significantly.(2)After exposure to BPA,the hierarchical structure of zebrafish retina and the arrangement of photoreceptor layer cells were disordered.(3)After the exposure to BPA and at the 24 hour post-fertilization(hpf),the number of zebrafish embryo spontaneous movement decreased,and the eye frequency and amplitude of juveniles at 96 and 120 hpf were reduced.The autonomous exercise ability and swimming distance of juvenile fish at 120 and 144 hpf were significantly reduced.Conclusion:Embryonic BPA exposure can cause the eye structural changes and abnormal visual behavior of zebrafish,which indicates that environmental pollutants may affect the development of offspring’s vision,and that children’s eye care should be conducted from the prenatal period.
