摘 要:目的:探究薰衣草总黄酮抗小鼠皮肤光损伤的有效性及氧化应激的相关作用机制,为薰衣草的开发利用提供依据。方法:将84只雌性昆明小鼠随机分为7组,正常组、UVB组、溶剂组、维生素E组(0.013 g/kg)、薰衣草总黄酮低、中、高剂量组(0.25、1.25、2.50 g/kg)。建立小鼠皮肤光损伤模型,观察薰衣草总黄酮对光损伤模型小鼠行为特征、背部皮肤状态的影响;采用苏木素-伊红和维多利亚蓝染色观察其组织病理学改变及小鼠表皮厚度的变化;Western blot法检测小鼠Nrf2信号通路相关蛋白的表达。结果:薰衣草总黄酮能够明显减轻小鼠皮肤外观损伤程度、表皮厚度及皮肤组织的病理学变化,且能显著增强Nrf2、Keap1、HO-1、NQO1、GCLC蛋白的相对表达。结论:薰衣草总黄酮对小鼠皮肤光损伤具有一定的防护作用,其作用机制可能与激活Nrf2信号通路、调控氧化应激反应有关。Objective:To explore the effect of total flavonoids of lavender on skin light damaged mice and related mechanism of oxidative stress to provide basis for the development and utilization of lavender.Methods:A total of 84 female Kunming mice were randomly divided into 7 groups:blank group,model group,UVB group,vitamin E group(0.013 g/kg),lavender total flavonoids low,medium and high dose groups(0.25,1.25,2.50 g/kg).The photodamaged mice model was established,and the effects of lavender total flavonoids on behavior characteristics and skin appearance of mice were observed;Hematoxylin eosin and Victoria blue staining were used to investigate the histopathological and epidermal thickness changes in mice;The expression of proteins related to oxidative stress pathway in mice was detected by Western blot.Results:Total flavonoids of lavender could significantly reduce damage degree,epidermal thickness and pathological changes of skin tissue,and significantly enhance the relative expression of Nrf2,Keap1,HO-1,NQO1 and GCLC proteins.Conclusion:Lavender total flavonoids had a certain protective effect on skin photodamage in mice,and its mechanism might be related to activating Nrf2 signal pathway and regulating oxidative stress response.
